Bcl2 enhances survival of newborn neurons in the normal and ischemic hippocampus

Abstract: Neuronal progenitors in the adult hippocampus continually proliferate and differentiate to the neuronal lineage, and ischemic insult promotes hippocampal neurogenesis. However, newborn neurons show a progressive reduction in numbers during the initial few weeks, therefore, enhanced survival of newborn neurons seems to be essential for therapeutic strategy. Bcl-2 is a crucial regulator of programmed cell death in CNS development and in apoptotic and necrotic cell death. Therefore, we tested whether Bcl-2 overex… Show more

“…Bcl-2 plays a protective role against apoptotic and necrotic cell death in response to various stimuli including ischemia (Sasaki et al, 2006;Zhang et al, 2006). It is known that HDACIs increase the expression of Bcl-2 (Chen et al, 1999;Faraco et al, 2006).…”

“…Structurally, bcl2l2 is a member of the bcl2 family, and bcl2 is known to enhance survival of newborn neurons in the hippocampus (26). Similarly, bcl2l2 has been shown to play a role in cell survival in cell lines (27).…”

“…10 In brief, sections were treated with 50% formamide and 2ϫ saline-sodium citrate buffer and then incubated in 2N HCl. After a wash, sections were incubated with rat monoclonal anti-BrdU antibody (1:100, Harlan Sera-Laboratory, Loughborough, UK) at 4°C overnight.…”
