Background:Posterior uveitis is a polyetiologic group of diseases with polymorphic clinical manifestations and a prolonged, chronic and recurrent course. In addition, it is commonly bilateral and often causes complications, loss of working capacity and early visual disability in young adults. It is an important task of current clinical ophthalmology to find biomarkers indicating the severity of inflammation and the likelihood of developing a recurrence of the initial process. Purpose: To determine the sensitivity of peripheral blood lymphocytes to adrenaline and acetylcholine in patients with primary and recurrent uveitis at different phases of the disease. Material and Methods: One hundred patients with idiopathic posterior uveitis were examined at different phases of the disease. The control group was composed of 16 healthy individuals of a similar age. The specific sensitivity of lymphocytes to neuromediators, adrenaline and acetylcholine (expression of T cell adrenergic and acetylcholine receptors) was assessed using our complex methodology (in conjunction with a parallel sampling method) for assessing the individual's sensitivity to medicaments (biological regulators) which has been developed at Immunology laboratory, Filatov Institute of Eye Disease and Tissue Therapy. The method involves obtaining lymphocytes from an individual, culturing lymphocytes with examined drugs immunohistochemically, and use of a peroxidase anti-peroxidase method with monoclonal T-cell antibodies. Results: Absolute and relative adrenoreceptor and acetylcholine receptor expression levels in patients with posterior uveitis were 1.8-2.0 times and 1.6-2.4 times, respectively, higher in patients with posterior uveitis compared to controls. Absolute adrenoreceptor and acetylcholine receptor expression values were 12.6% and 31.7%, respectively, higher in the recurrent process during active inflammation compared to the period of remission. However, in patients with posterior uveitis, adrenoreceptor and acetylcholine receptor expression values in the period of remission were still higher than normal values. The odds of an increased relative adrenoreceptor expression and the odds of an increased relative acetylcholine receptor expression on peripheral blood lymphocytes were 67.5-fold higher and 23.6-fold higher, respectively, among patients with posterior uveitis compared to controls. The correlation of adrenoreceptor expression and acetylcholine receptor expression with absolute counts of T cells and T helpers was stronger than the correlation with B cells, which reflects to a larger extent the association with the cellular immunity and to a lesser extent the association with the humoral immunity. There was a moderate correlation of adrenoreceptor expression and acetylcholine receptor expression with cell subsets of markers of early lymphocyte activation CD5, autoimmune aggression CD25, intercellular adhesion CD54 and apoptosis CD95 (Fas/APO-1). Immunoregulatory potential of pregnancy-specific β1-glycoprotein

The embryo, being half an antigenically “foreign” organism, should elicit a maternal immune response. During evolution, however, the mechanisms ensuring successful development of pregnancy have been formed. In particular, among factors providing immune tolerance during pregnancy are some proteins associated with pregnancy. The pregnancy-specific β 1-glycoprotein (PSG, PSG1; SP1; PSβG1) is a dominant fetoplacental protein produced by cyto- and syncytiotrophoblast cells, and it exhibits immunosuppressive properties. Our team of authors possesses a patented method for obtaining native human PSG preparation from blood serum of pregnant women, a mixture of PSG1, PSG3, PSG7, PSG9, and their isoforms and precursors. This review presents an analysis of our results for the period from 2015 to 2020. We studied the immunoregu-latory effects of the obtained PSG preparation at concentrations comparable to those observed in pregnancy (1, 10, 100 |ag/mL). The study was performed with peripheral blood cells obtained from non-pregnant women. It was found that PSG significantly increased the percentage of adaptive Tregs in vitro, as well as expression of CTLA-4, GITR, and production of IL-10 by these cells. It has been shown that PSG has a stimulating effect upon indoleamine-2,3-dioxygenase (IDO) activity of peripheral blood monocytes. For Th17 cells, we have demonstrated that PSG can suppress differentiation and proliferation of these cells, along with reduced production of critical proinflammatory cytokines (IL-8, IL-10, IL-17, IFNγ, MCP-1, TNF α). As for the memory T cells, PSG suppressed CD25 expression and IL-2 production by them, along with simultaneous decreased expression of Gfi1, hnRNPLL genes, thus preventing the formation of the “mature” CD45R0 isoform. PSG has been shown to inhibit naive T cells’ conversion to the terminally differentiated effector subpopulation of helper T cells. When analyzing PSG effects upon cytokine profile of immunocompetent cells, it was found that the protein predominantly suppresses the Th1 cytokine production by the studied cell types, and regulates the Th2 cytokine production in divergent manner. The results obtained are consistent with general concept of immunosuppression during pregnancy. Thus, PSG could be one of the factors preventing formation and implementation of immune response to placental antigens.

Regulatory framework and methodological approaches to evaluation of immunological effects of vaccination against brucellosis are not established, and the degree of immunological post-vaccinal rearrangement is not yet developed. Due to leading role of cellular immunity in formation of immune protection against brucellosis, evaluation the cellular response in response to antigenic stimulation may be considered the most informative and objective approach to analysis of immune changes in the body during vaccination. In order to develop the most diagnostically informative methods for design of antigen-stimulation cell tests in vitro, a careful selection of a stimulating agent (antigen) is required, which should have a sufficient activating potential, thus providing specificity of reaction under in vitro conditions. The aim of the present study is to study the in vitro specific activity of a protein-polysaccharide antigenic complex from the Brucella abortus 19 BA strain (BrAg), and an opportunity of its application in order to assess the formation of post-vaccinal cellular immunity against brucellosis.The study was performed with white laboratory mice (n = 50) immunized with the Brucella abortus 19 BA strain. The control group (n = 50) consisted of laboratory mice that received a sterile saline solution in a volume of 0.5 ml. Blood samples were taken from immunized and control animals before vaccination, and 7, 14, 21, and 30 days after immunization. By means of flow cytometry, the activation molecules CD25, CD69, MHC II and CD95, expressed on T lymphocytes (CD3+CD69+, CD3+CD25+, CD3+CD95+, CD3+MHC+) were determined. To observe the development of immunity, the intensity of expression of T lymphocyte activation markers was calculated using the stimulation quotient. BrAg was used for specific in vitro stimulation of T lymphocytes. The liquid brucellosis allergen (brucellin) was used as an antigen for comparison, when studying opportunity of BrAg usage for assessing the postvaccinal immunity development.The following results were obtained: BrAg has pronounced specific activity, it did not cause non-specific in vitro reactions (activation) of T lymphocytes, thus enabling its application as a test antigen when evaluating development of adaptive vaccine immunity against brucella.Experimental testing of brucellosis antigen for carrying out the in vitro antigen-stimulated cellular reactions, aiming for evaluation of post-vaccinal immunity development against brucellosis, showed that the usage of BrAg promotes increase in diagnostic sensitivity of cellular reactions under in vitro experimental conditions. The applied experimental antigen is a quite promising tool for development of laboratory algorithms for brucellosis diagnostics, and assessment of actual vaccination efficiency in cohorts previously vaccinated against brucellosis.
