Basic Fibroblast Growth Factor Protects against Rotenone-Induced Dopaminergic Cell Death through Activation of Extracellular Signal-Regulated Kinases 1/2 and Phosphatidylinositol-3 Kinase Pathways

Abstract: Administration of rotenone to rats reproduces many features of Parkinson's disease, including dopaminergic neuron degeneration, and provides a useful model to study the pathogenesis of Parkinson's disease. However, the cell death mechanisms induced by rotenone and potential neuroprotective mechanisms against rotenone are not well defined. Here we report that rotenone-induced apoptosis in human dopaminergic SH-SY5Y cells is attenuated by pretreatment with several growth factors, most notably basic fibroblast gr… Show more

“…Chadi et al reported that bFGF promotes dopaminergic cell survival in human fetal tissue strands transplanted into immunosuppressed 6-OHDA-lesioned rats [10]. Another group showed that bFGF protects against rotenone-induced dopaminergic cell death [11]. In our previous study, we found that bFGF inhibits ER stress-induced neuronal cell apoptosis in spinal cord injury and brain ischemia/reperfusion injury [12,13].…”

“…2 and 3 show pie charts summarizing the functions of the common proteins and the functions of the common proteins with changes, respectively. Major classes of proteins included those related to chaperone, cytoskeleton, and ribosome, all of which have been implicated in rotenone-induced pathology (23,46,47). Notably a significant number of proteins listed in Table I were novel and without known functions.…”

“…Moreover, fibroblasts genetically engineered to produce FGF-2 and implanted into the striatum attenuate the damage caused by intrastriatal injection of 6-OHDA (Shults et al, 2000). More recently, FGF-2 was reported to protect against rotenone-induced cell death in both SH-SY5Y cells and cultured dopaminergic neurons (Hsuan et al, 2006).…”
