Barcoded Microchips for Biomolecular Assays

Abstract: Multiplexed assay of analytes is of great importance for clinical diagnostics and other analytical applications. Barcode-based bioassays with the ability to encode and decode may realize this goal in a straightforward and consistent manner. We present here a microfluidic barcoded chip containing several sets of microchannels with different widths, imitating the commonly used barcode. A single barcoded microchip can carry out tens of individual protein/nucleic acid assays (encode) and immediately yield all assa… Show more

“…In addition to PSA, we included DNA analogs of the microRNA sequences miR-30c and miR-141 based on literature demonstrating the utility of microRNA biomarkers in blood for detection of prostate cancer. [28,29] To detect this panel, we designed three nanoswitches with different loop sizes and created a multiplexed nanoswitch mixture and tested detection of all biomarkers in buffer (Figure 5c). We then spiked the biomarkers in 20% serum and showed detection of individual biomarkers (Figure 5c, lanes 2-4) In this study, we showed that DNA nanoswitch barcodes can be used to detect up to six biomarkers in a single assay, and even a mixture of protein, antibody and nucleic acids.…”

“…To realize accurate and multiplexed biochemical assays in an even better way, we develop a barcoded microchips for biomolecular assays according to barcode symbology specification (Figure 2c). [21] Using the barcode reader/smartphone, up to 20 samples can be detected simultaneous in a single barcoded chip.…”
