Balancing Trained Immunity with Persistent Immune Activation and the Risk of Simian Immunodeficiency Virus Infection in Infant Macaques Vaccinated with Attenuated Mycobacterium tuberculosis or Mycobacterium bovis BCG Vaccine

Abstract: Our goal is to develop a pediatric combination vaccine to protect the vulnerable infant population against human immunodeficiency virus type 1 (HIV-1) and tuberculosis (TB) infections. The vaccine consists of an auxotroph Mycobacterium tuberculosis strain that coexpresses HIV antigens. Utilizing an infant rhesus macaque model, we have previously shown that this attenuated M. tuberculosis (AMtb)-simian immunodeficiency virus (SIV) vaccine is immunogenic, and although the vaccine did not prevent oral SIV infecti… Show more

“…Mesenteric LN cell suspensions were stimulated with 10 g/2 ϫ 10 6 cells of SIV Gag or HIV Env peptide pools (NIH Reference and Reagent Program) for 6 h, with brefeldin A being added for the last 5 h. Negative-and positive-control cultures included medium only or cell stimulation cocktail, respectively. Cells were stained as previously described (20,34,69), and 300,000 events were acquired on an LSRII Fortessa instrument and analyzed using FlowJo version 10 software. Gates were based on FMO controls, and cytokine analyses were performed after Boolean gating.…”

“…Consistent with this idea, Jensen and colleagues hypothesize that vaccine-induced trained immunity led to increased susceptibility of infant macaques to SIV infection through enhanced activation of CD4 ϩ T cells (22). They used a replication-deficient M. tuberculosis strain (panCD, leuCD, and secA2 deletion mutant) expressing the SIV Gag and Env antigens (attenuated M. tuberculosis-simian immunodeficiency virus vaccine [AMTB-SIV]) as a dual-purpose SIV-M. tuberculosis vaccine.…”

“…Jensen et al (22) point out that these frequencies exceeded the antigen-specific CD4 ϩ T cell responses (both SIV and TB) observed in their previous studies; thus, the comparatively elevated activation states of the CD4 ϩ T cell populations seen in this study appear to be unrelated to vaccine specificity. As indicated above, some of the highest responses after challenge were observed in the BCG-immunized animals.…”

“…The relevance of sex as a biological variable and rigor of experimental approaches were emphasized. Important reports on these and other trending topics were published in CVI (8)(9)(10)(11)(12)(13)(14). In this rapidly expanding scientific environment, CVI remained committed to disseminating discoveries and scientific knowledge with high scholarly and editorial standards, in line with those of other ASM journals and following the tradition and excellence of the society itself.…”
