Bakuchiol suppresses proliferation of skin cancer cells by directly targeting Hck, Blk, and p38 MAP kinase

Abstract: Bakuchiol is a meroterpene present in the medicinal plant Psoralea corylifolia, which has been traditionally used in China, India, Japan and Korea for the treatment of premature ejaculation, knee pain, alopecia spermatorrhea, enuresis, backache, pollakiuria, vitiligo, callus, and psoriasis. Here, we report the chemopreventive properties of bakuchiol, which acts by inhibiting epidermal growth factor (EGF)-induced neoplastic cell transformation. Bakuchiol also decreased viability and inhibited anchorage-independ… Show more

“…Bak is a meroterpene found in the traditional Chinese herbal medicine Fructus psoraleae [ 8 ]. Kim et al [ 18 ] found that Bak exhibits potent anti-cancer activity by directly targeting Blk, Hck, and p38 MAPK. Shoji et al [ 19 ] reported that Bak produced enantiomer-selective anti-influenza A virus activity via a novel mechanism involving the host-cell response, which contributes to the development of novel approaches for the treatment of influenza.…”

“…After 24 h, A431 cells were selected with 2 μg/mL of puromycin. The shMock or shPRPK -transfected A431 cells were incubated for 10 days as described previously ( 53 ). Colony formation was observed and analyzed using a LEICA DM IRB microscope (Nikon Corporation) and the ImagePro Plus software (v.6.1) program (Media Cybernetics Inc., Rockville, MD).…”

“…Other members of SFKs could be involved in specific steps of skin cancer progression. Kim et al showed that the EGF-stimulated anchorage-independent growth of A431 human epidermoid carcinoma cells could be inhibited by blocking the kinase activity of Hck and Blk [110].…”
