Baicalein and wogonin inhibit collagen deposition in SHR and WKY cardiac fibroblast cultures

Abstract: In order to demonstrate the potential therapeutic effect of two flavonoids, Baicalein and Wogonin, on suppression of pathological myocardial fibrosis in hypertension, we investigated their in vitro effects on collagen expression in primary cultured cardiac fibroblasts isolated from neonatal normotensive (WKY) and hypertensive (SHR) rats. Our results showed that over-expression of collagen mRNA and protein induced in cardiac fibroblasts by angiotensin (AngII) could be attenuated significantly by both flavonoids… Show more

“…Baicalein and wogonin are two of the main active ingredients in the Scutellaria baicalensis Georgi. In the CFs, baicalein and wogonin treatment suppresses collagen I and collagen III expression stimulated by Ang II (Kong et al, 2010 ). They also reduce myocardial collagen volume fraction and inhibit myocardial collagen I and III mRNA expression in spontaneously hypertensive rats by mediating ERK and MMP-9 pathways (Kong et al, 2010 , 2011 ).…”

“…[6][7][8] Besides, wogonin could inhibit angiogenesis, suppress collagen deposition in cardiac fibroblasts and inhibit ischemic brain injury. [9][10][11][12] There is no evidence for vascular relaxation effect of wogonin. We describe here that wogonin, unlike oroxylin A, is an endothelium-and NO-independent vasodilatory flavonoid.…”

“…This compound has been shown to possess a variety of biological characteristics, including antioxidative and anti‐inflammatory effects, tumour growth inhibition and protection of ischaemia/reperfusion injury . It has been proven to attenuate fibrotic activities in cardiac fibroblasts via mitogen‐activated protein kinase signalling and to suppress hepatic stellate cell migration by inhibiting monocyte chemoattractant protein‐1 expression . However, the effect of baicalein on human dermal fibroblasts (normal or exuberant) and the potential molecular mechanisms of baicalein are still unknown.…”
