BAFF Blockade Attenuates Inflammatory Responses and Intestinal Barrier Dysfunction in a Murine Endotoxemia Model

Abstract: B cell-activating factor (BAFF) production is increased in septic patients. However, the specific role of BAFF in sepsis remains unknown. This study was designed to investigate the expression and function of BAFF in an experimental endotoxemia model and to identify the potential mechanisms. We established an endotoxemia mouse (6–8 weeks, 20–22 g) model by administering 30 mg/kg lipopolysaccharide (LPS). BAFF levels in the circulating system and organ tissues were measured 4 and 8 h after LPS injection. Surviva… Show more

“…Several inflammatory models suggest that BAFF exhibits pro-inflammatory effects. BAFF significantly promotes the secretion of serum IL-1β, TNF-α, IL-6 and other inflammatory factors in the LPS-induced sepsis model whereas neutralization of BAFF markedly inhibits pro-inflammatory factor release ( 30 ). In rheumatoid arthritis (RA) model, BAFF promoted B-cell survival and also secretion of pro-inflammatory factors IL-6 and IL-8 in mice, consistent with the result in synovial fibroblasts co-cultured with INF-γ ( 31 ).…”

“…Candida is one of the most common fungi of the GI tract; however, bacterial dysbiosis can cause Candida commensalism to become pathogenic, resulting in prolonged infections and Candidiasis ( Strati et al, 2016 ). Sepsis can lead to the impairment of the intestinal barrier structure and function due to inflammation, ischemia, and hypoxia ( Quan et al, 2020 ). Invasive C. albicans infection can further impair the intestinal barrier and lead to disseminated candidiasis, which disrupts the microflora of the gut ( Geng et al, 2020 ).…”

“…Severe burns cause a wide range of immune responses and extensive cell damage, which is considered a precursor to organ dysfunction. Then, excessive inflammatory reaction would destroy endothelial and intestinal epithelial cells and lead to their apoptosis, resulting in increased intestinal permeability and aggravating intestinal mucosal barrier dysfunction; these effects are followed by endotoxin and bacterial migration and further intestinal flora imbalance ( 31 ). As a result, sepsis in patients progressively develops and deteriorates.…”
