BAFF and BAFF-Receptor in B Cell Selection and Survival

Abstract: The BAFF-receptor (BAFFR) is encoded by the TNFRSF13C gene and is one of the main pro-survival receptors in B cells. Its function is impressively documented in humans by a homozygous deletion within exon 2, which leads to an almost complete block of B cell development at the stage of immature/transitional B cells. The resulting immunodeficiency is characterized by B-lymphopenia, agammaglobulinemia, and impaired humoral immune responses. However, different from mutations affecting pathway components coupled to … Show more

“…Each receptor exhibits a unique expression pattern on distinct B cell subsets, in part accounting for differential functional roles during an immune response. 28 29). 25 All three of the BAFF/APRIL receptors can be shed from the cell surface by the activity of γ-secretase (BCMA) or ADAM proteases (BAFF-R and TACI).…”

“…28 Importantly, constitutive cleavage of BCMA and TACI generates functional decoy receptors, whereas BAFF-R shedding does not appear to generate a decoy but rather regulates B cell survival via modulation of surface BAFF-R expression. 29…”

“…BAFF via BAFF-R plays a role in B cell selection and B cell survival [17,24,25]. CD8+ Tregs had no significant effect on BAFF-R expression on B cells, suggesting that CD8+ Tregs may not influence B cell survival.…”

“…( 48 ) Belimumab, a monoclonal antibody to BAFF that blocks BAFF–BAFFR signaling, has been shown to be effective for systemic lupus erythematosus and approved by the U.S. Food and Drug Administration and European Medicines Agency for human use. ( 49 ) VAY736, a monoclonal antibody to BAFFR, has been shown to be effective for primary Sjogren’s syndrome and is now under clinical trial (Table 6). Blockade of APRIL–BCMA signaling by a monoclonal antibody to APRIL was shown to be effective in murine model of systemic lupus erythematosus and immunoglobulin A nephropathy.…”
