Bacteriophage Lambda; Abortive Infection of Bacteria Lysogenic for Phage P2

Abstract: Abstract. The efficiency of plating of wild-type X on a host lysogenic for P2 is less than 10-6, and only a small number of infected cells produce progeny phage. Lambda can adsorb and inject its DNA normally in such cells; the DNA can circularize and is not nicked or degraded, but replication is severely impaired. Mutants of P2, which as prophages no longer interfere with X, have been isolated and found to be recessive to wild type, implying that P2 prophage codes for a diffusible product involved in X interfe… Show more

“…Prophage P2 expresses a gene, old, whose product is responsible for the inhibition of A growth (34). The action of old appears to be to introduce a general replication block in cells in which the function of RecBCD is impaired (20). Accordingly, P2 is unable to lysogenize a recB or recC mutant unless it bears a mutation in old, and A is unable to replicate in a P2 lysogen if it inactivates RecBCD by making Gam (20,34,42).…”

“…The answer may have to do with host range. In E. coli lysogenic for phage P2, A does not grow (20), while a A hybrid bearing a substitution of the P22 erf region for the A red region does (17). Prophage P2 expresses a gene, old, whose product is responsible for the inhibition of A growth (34).…”

“…The P2 old gene product interferes with the growth of λ phage and kills E. coli cells that lack the recBCD nuclease-helicase. 75 Induction of λ prophage in a P2 old C -lysogenic strain results in partial degradation of many tRNA molecules. 76 Purified Old protein has been reported to have 5 0 to 3 0 exonuclease activity on DNA, as well as an uncharacterized ribonuclease activity.…”
