Bacteriocin-like substance from Bacillus amyloliquefaciens shows remarkable inhibition of Acanthamoeba polyphaga

Abstract: The effectiveness of a bacteriocin-like substance (BLS) produced by Bacillus amyloliquefaciens was tested against Acanthamoeba polyphaga strains, and its cytotoxic potential on Vero cells was investigated. Amebicidal activity of the purified BLS was tested by plate bioassays with concentrations ranging from 12.5 to 6,400 AU mL(-1). Damage to A. pholyphaga cells was monitored using an inverted microscope and counted in a Fuchs-Rosenthal chamber after 24, 48, and 72 h. According to the results obtained, the BLS … Show more

“…The broad-spectrum antimicrobial activity exhibited by the studied isolates is in agreement with results reported for another B. amyloliquefaciens strain, LBM 5006 which produced inhibitory activity against both Gram-positive and Gram-negative bacteria (Lisboa et al 2006), phytopathogenic fungi (Benitez et al 2010) and the amoebae Acanthamoeba polyphaga (Benitez et al 2011). However, contrary to the present study, the antibacterial compound produced by strain LBM 5006 was attributed to a bacteriocin-like substance (BLS) of 5 kDA (Lisboa et al 2006), and the activity against the amoebae Acanthamoeba polyphaga was due to the production of another BLIS (Benitez et al 2011).…”

“…In recent years new strategies have been developed, which involve the use of various natural substances. There are several evidences of amoebicidal activity of essential oils obtained from Euphorbiaceae plant, [10], Pterocaulon polystachyum [11,12], Allium sativum extract [13], Propolis [14], and BLS (Bacteriocin Like Substances) [11,[15][16][17]. BLS are ribosomally synthesized antimicrobial peptides, generally small (<10 KDa), amphipathic and cationic peptide of variable length and structure.…”

“…Bacteriocins produced by LAB, recently, have attracted much attention because of their GRAS status and potential use as natural additives directly in food or in food-packaging [20,21], and as potential therapeutic agents [22]. Some peptides have a broad spectrum of activity against various microorganisms, comprising Gram-positive and Gram-negative bacteria, protozoa, yeast, viruses [15]. In the present study we have assessed "in vitro" the amoebicidal activity of three BLS produced by our isolated LAB against Acanthamoeba polyphaga, determining the cell viability or other morphological changes at different times, up to the end of the investigation (144 h).…”
