Bactericidal activity of ciprofloxacin, norfloxacin and ofloxacin in serum and urine after oral administration to healthy volunteers

Abstract: A 200 mg oral dose of ciprofloxacin, norfloxacin or ofloxacin was administered to six healthy male volunteers in a three way cross-over study in order to examine the kinetics in humans in relation to the bactericidal activity in serum and urine. Serum concentrations for ciprofloxacin were similar to norfloxacin and lower than ofloxacin. Despite this fact, ciprofloxacin showed the highest serum bactericidal titers compared to norfloxacin and ofloxacin when serum-resistant Escherichia coli C14 was used as a test… Show more

“…Zeiler et al determined the UBTs of ciprofloxacin, NOR, and ofloxacin under various conditions. They demonstrated that at a neutral pH the UBTs for all three quinolones increased compared to those at pH 5.6 (27).…”

“…Calculation of AUBCs from serial measurement of SBTs has been proposed to be a rational method to compare antibacterial drugs (1,16). A number of investigators have determined SBTs after administration of ofloxacin or ciprofloxacin (5, 9, 18, 24), and three investigations have directly compared the SBTs of ciprofloxacin and ofloxacin after single-dose intravenous or oral administration (12,25,28). The overall results of the present study are consistent with previous investigations in finding SBTs and AUBCs for E. coli and E. cloacae to be very high for both drugs, lower for S. aureus and P. aeruginosa, and poorest for S. pneumoniae.…”

“…In a number of other studies, various dosages were examined noncomparatively. Van Several studies (3,14,19,25,37) investigated the SBA following oral administration of ciprofloxacin at various doses (mainly 500 and 750 mg). The same trend of good-to-excellent activity against members of the family Enterobacteriaceae, mediocre activity against P. aeruginosa, and poor performance against S. aureus and other gram-positive organisms was observed.…”
