One cut-flower and 14 pot anthurium cultivars were screened for resistance to anthurium blight by spraying four isolates of Xanthomonas campestris pv. dieffenbachiae (McCulloch and Pirone) Dye onto leaf surfaces in replicated experiments. Varying degrees of resistance were observed among the 15 cultivars tested. The pot cultivars Julia and Gemini were the most resistant, while the cut-flower cultivar Hearts Desire was the most susceptible. Each cultivar displayed different degrees of resistance to individual isolates of the pathogen. The results of this research permit the selection of clones with greater resistance for use in breeding and also create a baseline for comparing resistance of newly developed cultivars.

A new semi-selective medium for the isolation of Xanthomonas axonopodis pv. dieffenbachiae, the etiological agent of anthurium bacterial blight . On anthurium, leaf symptoms start as water-soaked spots visible near the margins which often become surrounded by yellowish areas. The pathogen moves into vascular tissues (and infected stems turn dark brown), eventually leading to death of the plant. Blight symptoms can be observed on spathes. X. a. dieffenbachiae is able to infect a broad range of aroids and the strains display a complex host range specificity (Chase et al. 1992). AbstractAims: Xanthomonas axonopodis pv. dieffenbachiae causes anthurium blight, which is regarded as the most threatening disease for the anthurium industry worldwide. The bacterium is listed as a quarantine pathogen in several regions, including Europe. We evaluated the use of Neomycin-CephalexinTrimethoprime-pivMecillinam 4 (NCTM4) medium for its isolation. Methods and Results: A total of 104 bacterial strains were inoculated onto NCTM4 and on the previously published Cellobiose-Starch (CS) and EsculinTrehalose (ET) media. The strain collection included: the anthurium blight pathogen, Xanthomonas strains, for which false positive results are known to occur using serological identification-tests; other bacterial pathogens of anthurium; and representatives of bacteria that are commonly present in the anthurium phyllosphere. Media were evaluated following the ISO 16140 protocol for the validation of alternative methods. Conclusion: Growth of the anthurium blight pathogen was better on NCTM4 and ET media than on CS. NCTM4 provided a better repeatability. It also displayed a lower rate of false positive and false negative results when the pathogen was isolated from plant extracts. Significance and Impact of the Study: This study will lead to improved isolation protocols of the anthurium blight in official procedures. NCTM4 medium could also favourably be used in studies, which aim to further understanding of the biology and epidemiology of this pathogen.

One of the most devastating Xanthomonas diseases affecting the Anthurium cut flower industry worldwide is the bacterial blight caused by Xanthomonas axonopodis pv. dieffenbachiae (Xad). The disease can be spread through latently infected tissue-cultured plants that are used for the propagation of Anthurium worldwide. Current disease diagnostic techniques involve the use of semi-selective media and serological tests. This study describes the development of a PCR tool combined with a genus-specific monoclonal antibody for the sensitive detection of the pathogen directly from plants. It was demonstrated that the immunocapture PCR (IC-PCR) was more sensitive than the conventional PCR and even more sensitive than indirect ELISA for the detection of the pathogen. Latently infected plants could be positively screened for the presence of the pathogen. Three sets of primers were designed from DNA probes that were reported to show some specificity to the pathovar dieffenbachiae. The use of all three sets of primers in a single reaction successfully amplified the three individual loci when bacterial DNA was used as a template. The multiplex PCR generated PCR profiles that could differentiate between the reference strains of X. axonopodis pv. dieffenbachiae from other control bacteria. The new primers could therefore be used both for the diagnosis of Anthurium blight in single PCR reactions and also for the profiling of Xanthomonas. pv. dieffenbachiae strains using the multiplex PCR technique.
