Bacillus hisashii sp. nov., isolated from the caeca of gnotobiotic mice fed with thermophile-fermented compost

Abstract: A taxonomic study was performed on 15 bacterial isolates from the caeca of gnotobiotic mice that had been fed with thermophile-fermented compost. The 15 isolates were thermophilic, Gram-stain-positive, facultatively anaerobic, endospore-forming bacteria, and were most closely related to Bacillus thermoamylovorans CNCM I-1378 T isolate. All 15 isolates grew at 25-60 8C (optimum 50 8C), pH 6-8 (optimum pH 7) and were capable of growing on a medium containing 6 % (w/v) NaCl (optimum 0.5 %). The 15 isolates could … Show more

“…To construct the meta‐fermentation system for l ‐LA production, the isolated strains of W. coagulans (MN‐07, MC‐02, MN‐09, 24C‐1, NYC‐4, NA50‐3, and CPh‐14) and C. thermoamylovorans (MC‐07, OM55‐6, MC‐04, CPh‐4, and MN‐04) [ 18 ] were considered based on DGGE analysis. [ 7 ] Additionally, two strains (N‐11 and N‐21) of C. hisashii , which exhibits 99.4% pairwise sequence similarity with C. thermoamylovorans , [ 17 ] were investigated. C. hisashii and C. thermoamylovorans are not distinguishable based on 16S rRNA gene fragments in both DGGE and MiSeq 16S amplicon analyses.…”

“…Because the cell sizes are reported to be similar among W. coagulans , C. thermoamylovorans , and C. hisashii , [ 17,21,22 ] the cell concentrations would be proportional to the OD values of each bacterial culture. The pre‐cultured of each strain were individually performed for 20 h, and then, the respective pre‐cultured broths (OD 610 = 1.0) of 3 isolates were inoculated directly into 400 ml of MKR medium in a 1000‐ml jar fermenter equipped with a pH controller (DJ‐1023P, Able, Tokyo, Japan).…”
