Members of the genus Bacillus produced a diverse group of antimicrobial compounds. Here, we presented the antifungal activity and genome sequence analysis of Bacillus sp. 2211, a potential plant-growth-promoting bacterium. Bacterial supernatants from strain 2211 cultures in nutrient broth (NB) and potato dextrose broth (PDB) suppressed the mycelial growth of Pyricularia oryzae, Colletotrichum aenigma, Colletotrichum fructicola and Fusarium oxysporum. The supernatants were also able to suppress spore germination of these fungi, except for F. oxysporum. However, the supernatant from PDB displayed a significantly higher inhibition activity than NB. Additionally, the supernatant from PDB significantly reduced the disease severity caused by P. oryzae on rice seedlings. The genome of strain 2211 was sequenced. The highest digital DNA-DNA hybridization (80.1%) and average nucleotide identity (97.57%) levels indicated that strain 2211 was a member of the species Bacillus velezensis. The phylogenomic analysis showed that it clustered with B. velezensis NRRL B-41580T, B. velezensis KACC 13105 and B. velezensis subsp. plantarum FZB42T. The gene expression analysis showed the up-regulation of nonribosomal peptide synthetase (NRPS) genes bmyA, fenB and dhbE in PDB, compared to NB. This work demonstrated that the culture media affected the antagonistic activity of strain 2211 possibly through the modification of NRPS biosynthesis genes.

Application of the biosurfactant produced by Bacillus velezensis MMB‐51 as an efficient synergist of sweet potato foliar fertilizer

This study focused on the application of a microbial biosurfactant as a synergist of the foliar fertilizer. A biosurfactant‐producing Bacillus velezensis MMB‐51 was firstly isolated, which produced the peak yield of 2.287 g/L biosurfactant BS‐51 at the optimum fermentation condition. The chemical nature of BS‐51 was identified as lipopeptide consisting of surfactin and fengycin, which exhibited a low CMC of 11.0 mg/L in ddH2O and a good stability in wide ranges of temperature, pH and NaCl concentration. The BS‐51 was then applied as a synergist of three common sweet potato leaf fertilizers. The result showed that BS‐51 increased the contact area of fertilizer with leaf surface by 51%–93% and the moving speed of fertilizer on the inclined leaf surface by 272%–459%, indicating that BS‐51 decreased the surface tension of three‐leaf fertilizers. BS‐51 also increased the fertilizer residue after it slipped from the inclined leaf surface from 18.5%–21.5% to 29.4–31.2% depending on their types, thereby improving the utilization efficiency of foliar fertilizer. In addition, B. velezensis MMB‐51 secretion inhibited the growth of sweet potato pathogen Ceratocystis fimbriata. Therefore, B. velezensis MMB‐51 could act as an efficient synergist of crop foliar fertilizer with antifungal activity.

Cocultures have been widely explored for their use in deciphering microbial interaction and its impact on the metabolisms of the interacting microorganisms. In this work, we investigate, in different liquid coculture conditions, the compatibility of two microorganisms with the potential for the biocontrol of plant diseases: the fungus Trichoderma harzianum IHEM5437 and the bacterium Bacillus velezensis GA1 (a strong antifungal lipopeptide producing strain). While the Bacillus overgrew the Trichoderma in a rich medium due to its antifungal lipopeptide production, a drastically different trend was observed in a medium in which a nitrogen nutritional dependency was imposed. Indeed, in this minimum medium containing nitrate as the sole nitrogen source, cooperation between the bacterium and the fungus was established. This is reflected by the growth of both species as well as the inhibition of the expression of Bacillus genes encoding lipopeptide synthetases. Interestingly, the growth of the bacterium in the minimum medium was enabled by the amendment of the culture by the fungal supernatant, which, in this case, ensures a high production yield of lipopeptides. These results highlight, for the first time, that Trichoderma harzianum and Bacillus velezensis are able, in specific environmental conditions, to adapt their metabolisms in order to grow together.
