The Chinese revered a species of aquatic reptile known as Pelodiscus sinensis as both an edible and medicinal species. When artificially breeding, many deaths occurred at the farmed P. sinensis, mainly due to excessive breeding density, water contamination, and turtles biting each other secondary to bacterial infections. In this study, an isolate of gram‐negative bacteria WH0623 was isolated from the liver and kidney of diseased P. sinensis to trace the potential pathogen of this disease. Based on biochemical characteristics and 16S rRNA gene sequencing analyses, this isolated strain of WH0623 was identified as Chryseobacterium indologenes. The strain's median lethal dose (LD50) was 3.3 × 105 colony‐forming units (CFU)/g per fish weight tested using artificial infection. Histopathological analysis revealed pathological changes, including cell swelling, hyperaemia, and necrosis in many tissues. Antibiotic susceptibility tests revealed that the bacteria WH0623 was susceptible to doxycycline, sulphonamides, ceftazidime, norfloxacin, and ciprofloxacin. These antibiotics could treat the disease. In conclusion, the pathogen causing the death of farmed P. sinensis was isolated and identified, and a drug‐sensitive test was conducted. Our findings contribute to the future diagnosis and treatment of the disease.

The cuttlefish Sepia pharaonis is rich in nutritional value, and thus has been cultured in large-scale artificial culture. During its culture process in 2021 and 2022, a contagious disease with a mortality as high as 90% occurred when the dorsal mantle length of the cuttlefish was approximately 2.5 cm, which presented as obvious symptoms of small white spots on their dorsal mantle surface. To address the cause of the disease outbreak from a microbiome perspective, the bacterial communities in the healthy, infected, and dead cuttlefish S. pharaonis were assessed using bacterial culturing and 16S rRNA sequencing technology. Cultivable bacteria showed that there was an antagonistic phenomenon in the healthy S. pharaonis, and the highest diversity of bacteria was found in the dead individuals. The bacterial community was distinctly different among the three groups of samples, characterized by decreased alpha diversity and alteration of the dominant bacteria and bacterial community structures as the disease progressed. In addition, bacterial genera can be used as biomarkers for this disease, including Photobacterium, Bacillus, and Halarcobacter. In conclusion, the balance of the microbiome is necessary for S. pharaonis to remain healthy, and biomarker bacteria can aid in the early detection of occurrences of disease.

Chinese soft‐shelled turtle Pelodiscus sinensis possesses a distinctively soft and pliable apron around its dorsal carapace termed calipash, which is of great importance to evaluate the nutritional quality in turtle industry. As bacterial infection such as Aeromonas hydrophila often brings severe economic losses to P. sinensis culturing in China, it is necessary to study the response of this most distinctive calipash tissue to bacterial infection, which has not been reported. In this study, the texture quality, histology and extracellular matrix‐related molecules of calipash in P. sinensis after A. hydrophila challenge were evaluated. After A. hydrophila infection, calipash tissue samples were subjected to biological evaluation, histological analyses using hematoxylin and eosin staining, Masson trichrome staining, scanning electron microscopy and real‐time polymerase chain reaction for measuring mRNA expression of collagen biosynthesis as well as collagen degradation‐related genes. Picrosirius red staining was used to reveal the type I collagen distribution. The results showed histologically observable connective tissue disorganization with a notable increase in collagen‐characteristic amino acid hydroxyproline level, greater collagen I density and more collagen content in infected turtles when compared to the control. For the infected, significantly decreased mRNA levels of type I collagen and proline‐4‐hydroxylase coding genes, increased matrix metalloproteinases and lowered tissue inhibitor of metalloproteinase expression were observed. These results suggest that collagen metabolism or mobilization characterized using collagen degradation may play an important role in the maintenance of calipash in response to the A. hydrophila challenge.
