Corrole Nanoparticles for Chemotherapy of Castration-Resistant Prostate Cancer and as Sonodynamic Agents for Pancreatic Cancer Treatment

Abstract: A nanoparticle-based system, composed of the gallium(III) complex of a minimally substituted corrole that is coated by transferrin as a targeting vehicle (3-Ga NPs), has been used for pre-clinical evaluation of its efficacy against human metastatic castration-resistant prostate cancer (mCRPC) tumor xenografts. All mice (N = 9) responded to a dose of 10 mg/kg, with a remarkable tumor growth inhibition of 400% following 2 weeks of treatment; Ames and hERG tests excluded potential concerns regarding mutagenicity … Show more

“…This was accompanied by the targeted induction of tumor cell necrosis through plasma membrane destabilization; the impairment of calcium homeostasis, lysosomal stress, and rupture; and the formation of reactive oxygen species (ROS). The in vivo relevance was addressed by using such NPs for the chemotherapy of castration-resistant prostate cancer as well as for much more difficult to treat pancreatic cancer . Chemotherapy was insufficient in the latter case, but this was resolved via sonodynamic therapy (SDT), taking advantage of earlier research that exposed corroles as potential agents for inducing the formation of singlet oxygen via sonication …”

“…However, there is no general agreement of such a mechanism, and in most cases, singlet oxygen has been observed as the main contributors among all other types of reactive oxygen species (ROS) . In most of the applications, molecular sonosensitizers are combined with microbubbles or other sonosensitizer nanoparticles to enhance the therapeutic performance. …”
