B-cell activity markers are associated with different disease activity domains in primary Sjögren’s syndrome

Abstract: ObjectivesB-cell activating factor (BAFF), β-2 microglobulin (β2M) and serum free light chains (FLCs) are elevated in primary SS (pSS) and associated with disease activity. We aimed to investigate their association with the individual disease activity domains of the EULAR Sjögren’s Syndrome Disease Activity Index (ESSDAI) in a large well-characterized pSS cohort.MethodsSera from pSS patients enrolled in the UK Primary Sjögren’s Syndrome Registry (UKPSSR) (n = 553) and healthy controls (n = 286) were analysed f… Show more

“…As α 1 -MG is not an acute phase protein, variations in its urinary levels can be safely inferred not to be caused by a transient state of systemic inflammation (30). While FLC and β2M testing may also be associated with renal damage, their role as diagnostic biomarkers was not considered yet (26).…”

“…Area under the curve was best for the combination of creatinine and α 1 -MG, with a significant difference compared to creatinine alone (0.824 vs. 0.777) (24). In 2018, a study was published in which B-cell activity markers and organ involvement in pSS were considered (26). Levels of serum free light chains (FLC) and β2-microglobulin (β2M) were found to be associated with renal disease.…”

“…Anti-Ro/SSA, anti-La/SSB, RF, IgG, immunoglobulin A (IgA), immunoglobulin M (IgM), C3, C4, ESR, white blood cell count (WCC) and C-reactive protein (CRP) were measured by the clinical laboratory of the recruiting hospitals. BAFF, β2 microglobulin (B2M), κ light chain, λ light chain, κ-λ ratio, and combined free light chains (CMBY) were measured by The Binding Site (Birmingham, UK) as previously described [24].…”

“…However, we are unaware of studies that focus on the effects of dysregulation of transcriptomes, including lncRNAs, of B cells on the pathogenesis of pSS. Serum level of B cell-activating factor was associated with disease activity of pSS [16][17][18]. In addition, the distribution of peripheral B cell subsets is profoundly altered in patients with pSS [19].…”
