Aβ Secretion and Plaque Formation Depend on Autophagy

Abstract: Alzheimer's disease (AD) is a neurodegenerative disease biochemically characterized by aberrant protein aggregation, including amyloid beta (Aβ) peptide accumulation. Protein aggregates in the cell are cleared by autophagy, a mechanism impaired in AD. To investigate the role of autophagy in Aβ pathology in vivo, we crossed amyloid precursor protein (APP) transgenic mice with mice lacking autophagy in excitatory forebrain neurons obtained by conditional knockout of autophagy-related protein 7. Remarkably, autop… Show more

“…These accumulated autophagic vacuoles potentially provide membrane structures to produce A␤, as BACE1-cleaved APP and ␥-secretase components are enriched in autophagic vacuoles (57). Moreover, impaired autophagy was reported to promote the intracellular accumulation of A␤ and subsequent neuronal degeneration (58). Thus, inhibition of p38␣ MAPK FIGURE 6. p38␣ MAPK deficiency does not change the phosphorylation of BACE1 in SH-SY5Y cells.…”

“…Recent reports have shown that autophagy plays a role in unconventional protein secretion in mammalian cells. 11,13,14,22 To examine whether Ab-induced IDE secretion is associated with autophagy, we utilized several autophagic modulators. We found that Ab-induced autophagy is activated in astrocytes ( Fig.…”

“…Autophagic degradation of tau is regulated by nuclear factor erythroid-2-related factor 2 (Nrf2)-mediated induction of the autophagy receptor NDP52 (Jo et al, 2014) and the Alzheimer's-disease-associated locus phosphatidylinositol binding clathrin assembly protein (PICALM, also known as CALM), which controls the endocytosis of soluble NSF attachment protein receptors (SNAREs) that are involved in the autophagy pathway . Autophagy also regulates amyloid-b production , and defective autophagy leads to the accumulation of intraneuronal amyloid precursor protein (APP) (Nilsson et al, 2013). In microglia, amyloid-b is degraded by autophagy through the autophagy receptor optineurin (Cho et al, 2014) (see poster).…”
