Invasion, cell proliferation and apoptosis are important biological features of neoplasia, bearing prognostic importance. Histological stage, mitotic index, and apoptotic index have been assessed in 33 feline malignant mammary tumors. Histological stage (P < 0.01) and mitotic index (P < 0.001) had a significant association with prognosis in univariate analysis. Apoptotic index did not correlate with survival (P = 0.44), and histological stage (P = 0.48) did not correlate with mitotic index (P = 0.39). In feline malignant mammary tumors the apoptotic index seems unable to predict survival and lacks any correlation with proliferation assessed as mitotic index. A possible explanation for the lack of correlation between apoptotic index and survival may be due to the rapid acquisition of pathways of apoptosis resistance in feline mammary tumors or to rapid hormone receptors loss.

Mitotic index is a clinically important parameter in cancer pathology. We developed a staining method using Toluidine Blue to detect efficiently and rapidly mitotic figures in sections of formalin-fixed paraffin-embedded human and rat tissues. Sections were stained at acid pH with a 0.01% Toluidine Blue solution after removal of RNA with hydrochloric acid or ribonuclease. The optimal pH of the TB staining solution was found to be 4.5 for rat tissues and 3.5 for human tissues. This procedure stained mitotic figures much more intensely than other (extra)cellular structures. A quantitative estimate of the total number of nuclei in the field where mitotic figures were counted, was obtained in an adjacent section hydrolysed in 5 N hydrochloric acid and stained by the Feulgen reaction with a Schiff-type reagent containing 0.01% Toluidine Blue. This method specifically stained interphase and mitotic nuclei and the field cellularity could be quantified by image cytometry. When these procedures were performed on two consecutive serial sections, a mitotic index could be determined accurately by relating the count of mitotic figures to the number of tumour cells.

Citrus vein‐enation virus‐induced tumors on leaves, thorns, stems, and roots of Citrus auranti‐folia (Christm.) Swingle and C. jambhiri Lush. were investigated. Leaf vein tumors are initiated as cytological abnormalities of phloem fiber primordial cells adjacent to protophloem sieve tubes. The tumors then enlarge through hyperplasia of the affected fiber primordia. The mesophyll and epidermal tissues on the abaxial side of affected veins divide less prolifically and contribute to the tumor mass. Leaf vein tumors are determinate, like the protophloem where they originate, and cease enlarging as the leaf matures. Most thorn tumors are initiated in fiber primordia and develop similarly to vein tumors. Stem tumors and some thorn tumors develop from affected cells of the procambial tissue that lies between the metaxylem and metaphloem of vascular bundles. The cambium which differentiates in these areas is composed of affected cells and produces large amounts of abnormal xylem tissue. The resulting woody tumors are indeterminate like the cambium and secondary xylem.
