Avian Immune Capacity and Bone Marrow Cellularity after in ovo Treatment with Cyclophosphamide

Abstract: Lymphoid tissue, immunity and erythropoiesis in bone marrow were studied in chickens which had received air cell applications of cyclophosphamide (Cy) between 16 and 18 days of their embryonic development. The bursae from the Cy-treated birds were reduced significantly in size, deficient in bursal follicles, and lacked lymphocytes. The agglutinin level of sheep red blood cells of birds treated with Cy (2 mg) as 16, 17 and 18-day embryos was significantly lower than controls. While these Cy-treated birds lacked… Show more

“…Cyclophosphamide has been used experimentally in poultry as an immunosuppressive agent (Lerman and Weidanz, 1970;Glick, 1971Glick, , 1986Linna et al, 1972;Rouse and Szenberg, 1974;Sharma and Lee, 1977;Nathanson et al, 1981;Fadly et al, 1985;Ficken and Barnes, 1988;Corrier et al, 1991). However, at the dosages used, CPA lacks specificity in birds because lymphocytes, thrombocytes, and heterophils have all been shown to be affected (Lerman and Weidanz, 1970;Glick, 1971Glick, , 1986Linna et al, 1972;Rouse and Szenberg, 1974;Sharma and Lee, 1977;Nathanson et al, 1981;Fadly et al, 1985;Ficken and Barnes, 1988). In mice, 5-FU at specific doses has been shown to have a dramatic and specific effect on the circulating neutrophil population (Moore and Warren, 1987;Shimamura et al, 1987;Mclntyre et al, 1989).…”

“…5 Because of the low relative hatch of SB eggs (12%) various concentrations of Cy were used to determine the highest dose of Cy that would not significantly influence mortality. Cyclophosphamide solutions of 0, .5, 1.0, 1.5, and 2.0% were aseptically applied to the air cell of SB eggs at Days 16,17, and 18 of embryonic development (Glick, 1986).…”
