-The Brazilian pastures establishment success depends on the use of seeds with high physiological and sanitary quality. The aim of this study was to evaluate the effect of thermal and chemical treatments on quality of Brachiaria brizantha, cv. BRS Piatã and pathogen transmission via seed. The treatments included the use of fungicides, detergent, sodium hypochlorite, sulfuric acid and alcohol, as well as thermal treatments. In addition to seeds without treatment, nine treatments were tested.The tests used to assess the physiological quality of the seeds were first and final count of germination test and root protrusion; germination and root protrusion speed index. A transmitting test and filter paper method had been performed for sanitary quality. Fungal lower incidence was observed in seeds treated with alcohol, however, this treatment reduced the physiological quality of the seeds and higher frequency of fungi transmission from seeds to seedlings was observed for Bipolaris sp., Fusarium sp. and Phoma sp. Treatment with sulfuric acid provided a better seed germination performance and reduced the incidence of fungi.Index terms: germination, vigor, seed-seedling, incidence, forage.Qualidade fisiológica, sanitária e transmissão de fungos associados às sementes de Brachiaria brizantha (Hochst. ex. A. Rich.) Stapf submetidas a tratamentos térmicos e químicos RESUMO -O sucesso no estabelecimento das pastagens brasileiras depende da utilização de sementes com boa qualidade fisiológica e sanitária. Objetivou-se com o trabalho avaliar o efeito de tratamentos térmicos e químicos sobre a qualidade fisiológica e sanitária de sementes de Brachiaria brizantha, cultivar BRS Piatã e a transmissão de patógenos via semente. Os tratamentos incluíram o uso de fungicidas, detergente, hipoclorito de sódio, álcool e ácido sulfúrico, além de tratamentos térmicos. Os testes utilizados para a avaliação da qualidade fisiológica das sementes foram primeira contagem e contagem final do teste de germinação e de protrusão radicular; índice de velocidade de germinação e de protrusão radicular. Para a qualidade sanitária foi realizado o método de papel de filtro e o teste de transmissão. Menor incidência fúngica foi observada nas sementes tratadas com álcool, no entanto, esse tratamento reduziu a qualidade fisiológica das sementes e maior frequência de transmissão de fungos das sementes para as plântulas foi observada para Bipolaris sp., Fusarium sp. e Phoma sp. O tratamento com ácido sulfúrico proporcionou melhor desempenho germinativo das sementes e reduziu a incidência de fungos.Termos para indexação: germinação, vigor, semente-plântula, incidência, forrageira.

Resumo -O objetivo deste trabalho foi avaliar os padrões eletroforéticos das proteínas resistentes ao calor e a atividade da enzima endo-β-mananase durante a germinação de sementes de alface, em alta temperatura. Sementes de oito cultivares de alface foram submetidas aos testes de germinação, primeira contagem e emergência em duas temperaturas, 20 e 35 o C. Foram calculados o índice de velocidade de germinação (IVG) e o índice de velocidade de emergência (IVE). Avaliou-se, também, a expressão das proteínas resistentes ao calor e da enzima endo-β-mananase, para todos os tratamentos. Utilizou-se o delineamento experimental inteiramente casualizado, em arranjo fatorial 2x8, com duas temperaturas e oito cultivares. A maior germinação é observada a 35 o C na cultivar Everglades, considerada termotolerante. Os padrões de proteínas resistentes ao calor em sementes de alface apresentam bandas específicas na cultivar Everglades, a 35ºC. A atividade da enzima endo-β-mananase é maior na cultivar Everglades, nessa temperatura. Essa cultivar tem potencial para utilização em programas de melhoramento de alface com vistas à tolerância a altas temperaturas durante a germinação.Termos para indexação: Lactuca sativa, endo-β-mananase, proteínas tolerantes ao calor, termoinibição. Physiological and biochemical aspects of lettuce seed germination at different temperaturesAbstract -The objective of this work was to evaluate the electrophoretic patterns of heat-tolerant proteins and the activity of the endo-β-mannanase enzyme during lettuce seed germination, at high temperature. Seeds of eight lettuce cultivars were subjected to germination, first count, and emergence tests at two temperatures, 20 and 35°C. The index of germination speed (IVG) and the emergence rate index (IVE) were calculated. The expression of heat-tolerant proteins and of the endo-β-mannanase enzyme was also evaluated for all treatments. The experimental design was completely randomized, in a 2x8 factorial arrangement, with two temperatures and eight cultivars. The highest germination is observed at 35°C in the Everglades cultivar, considered thermotolerant. The patterns of heat-tolerant proteins in lettuce seeds present specific bands in the Everglades cultivar, at 35ºC. The activity of the endo-β-mannanase enzyme is higher in the Everglades cultivar, at this temperature. This cultivar has potential for use in lettuce breeding programs aiming for tolerance to high temperatures during germination.

The evaluation of seed physiological quality can help taking quick decision about the use or disposal of lots for conservation. The tetrazolium test is one of the most traditional in the analysis of seeds quality. Optional preconditioning procedures have improved the efficiency of this test. This work compared the efficiency of different preconditioning procedures, the buffered solution, concentrations and exposure time to tetrazolium solution in assessing the quality of orchid seeds of Cattleya labiata and Cattleya tigrina lots. The analysis was made through magnification of digital images. For preconditioning evaluation, two seed lots were aliquoted and submitted to different treatments: 10% sucrose solution, 10% glucose solution and water at room temperature for 24 hours. After preconditioning, seeds were immersed in tetrazolium solutions -prepared with buffer or water -at concentrations of 0.1, 0.5 and 1.0% for periods of 3, 12 and 24 hours, at 40 °C, in a water bath, in the dark. The experiment design was arranged in a factorial 4x3x3x2 and the mean comparison was made by Tukey test at 5% probability. Tetrazolium test in preconditioned orchid seeds allowed better visualization of red viable seeds. Tetrazolium test in orchid seeds preconditioned in glucose 10% solution or sucrose 10% solution was more effective for viable seeds visualization. Diluting tetrazolium salt in buffered solution or water made no difference for the results. Maintaining seeds in contact with tetrazolium 1% solution for 24 hours provided the higher viability values for both seed lots.
