Autophagy in Viral Development and Progression of Cancer

Abstract: Autophagy is a complex degradative process by which eukaryotic cells capture cytoplasmic components for subsequent degradation through lysosomal hydrolases. Although this catabolic process can be triggered by a great variety of stimuli, action in cells varies according to cellular context. Autophagy has been previously linked to disease development modulation, including cancer. Autophagy helps suppress cancer cell advancement in tumor transformation early stages, while promoting proliferation and metastasis in… Show more

“…The role of autophagy in pathogenic processes and antitumour therapy has been an issue of concern in recent years [20][21][22]. The autophagic process of tumour cells was found to be an important survivaland growth-associated mechanism in many cancers in a previous study [23][24][25].…”

“…Viruses, including coronaviruses, suppress autophagy to promote their own replication, and consequently activation of autophagy can effectively limit viral replication. [69][70][71][72] SARS-CoV-2 blocks autophagy, leading to the suggestion that autophagy-inducing agents may overcome infection by reactivating intracellular viral destruction. [73][74][75][76][77][78][79] Supporting this concept, SARS-CoV-2 was shown to promote Beclin-1 degradation, 80 and several autophagy-modifying compounds suppress SAR-CoV-2 replication in vitro (reviewed in 75 ).…”

“…Also, it is crucial to maintain homeostasis and avoiding from nutritional, metabolic, and infection-mediated stresses [ 91 ]. Moreover, its role in diseases progression especially cancers in different stages of them has been discussed previously [ 92 , 93 ]. Also, autophagy plays important role in viral replication and coordinates adaptive immunity by delivering virus-derived antigens for presentation to T lymphocytes [ 94 ].…”
