Autophagy in cardiovascular diseases: role of noncoding RNAs

Abstract: Cardiovascular diseases (CVDs) remain the world’s leading cause of death. Cardiomyocyte autophagy helps maintain normal metabolism and functioning of the heart. Importantly, mounting evidence has revealed that autophagy plays a dual role in CVD pathology. Under physiological conditions, moderate autophagy maintains cell metabolic balance by degrading and recycling damaged organelles and proteins, and it promotes myocardial survival, but excessive or insufficient autophagy is equally deleterious and contributes… Show more

“…In addition, the recent clinical relevance of ncRNAs in CVDs has been discussed in detail in a published report. 149 Although the overall understanding of ncRNAs is in its infancy, it is believed that with continuous exploration of the mechanism by which ncRNAs regulate pyroptotic pathways, ncRNAs could serve as a promising therapeutic strategy for pyroptosis-related CVDs.…”

“… 81 Additionally, miRNAs and long noncoding RNAs (lncRNAs) can be regulated by autophagy, including plasmacytoma variant translocation 1 (PVT1). 82 , 83 Although the mechanism is unknown, the elevation of PVT1 mediated by autophagy has an important function in the development and progression of diabetic nephropathy. 82 , 84 As another important type of noncoding RNA, circRNA expression is also most likely regulated by autophagy, at least during its back-splicing generation process, which may result in the impairment of circRNA functions in human physiology and pathology, including cancer.…”

“…Although increasing attention has been put on exploring the therapeutic potential of miRNA to treat CVDs, not enough research has been done on other ncRNA types, such as lncRNA. LncRNA has been closely associated with pathways, such as autophagy regulation, proliferation, apoptosis [ 160 ]—all of which have implications in CVDs, indicate the plausible role of lncRNAs in CVDs. In order to establish an association between lncRNAs and CVDs, we have previously demonstrated the differential regulation of several lncRNAs in ECs following treatment with atherosclerosis-associated oxLDL and pravastatin [ 161 ]; diabetes-associated high glucose [ 162 ]; inflammation-associated lipopolysaccharides; and fibrosis-associated TGFβ [ 163 ].…”
