Autophagy in cancer: a complex relationship

Abstract: Macroautophagy is the process by which cells package and degrade cytosolic components, and recycle the breakdown products for future use. Since its initial description by Christian de Duve in the 1960s, significant progress has been made in understanding the mechanisms that underlie this vital cellular process and its specificity. Furthermore, macroautophagy is linked to pathologic conditions such as cancer and is being studied as a therapeutic target. In this review, we will explore the connections between au… Show more

“…In 2011, the idea of “autophagy addiction” was proposed as a promising therapeutic approach for certain tumours . In reality, autophagy plays different roles in cancer biology, depending on the tumour type and the context . In OSCC, various studies have drawn inconsistent conclusions regarding the associations between the levels of autophagy‐related proteins and a poor prognosis …”

“…6 In reality, autophagy plays different roles in cancer biology, depending on the tumour type and the context. 7 In OSCC, various studies have drawn inconsistent conclusions regarding the associations between the levels of autophagy-related proteins and a poor prognosis. 8,9 Autophagy initiation is controlled through unc-51-like kinase-1 (ULK1) and vacuolar protein sorting-34 (VPS34).…”

“…Autophagy describes the fundamental catabolic mechanism during which cells degrade dysfunctional and unnecessary cellular components (41,42). This process is driven by the action of lysosomes and promotes survival during starvation periods, as the cellular energy level can thus be maintained (42). In this study, RSF did not induce the autophagy (data not shown), but the apoptosis.…”

“…Apoptosis involves a complex cascade of reactions regulated by specific proteases called caspases, and results in DNA degradation (34).
