Autonomic cardiocirculatory control in mice with reduced expression of the vesicular acetylcholine transporter

Abstract: In cardiovascular diseases, sympathetic tone has been comprehensively studied, whereas parasympathetic tone has received minor attention. The vesicular ACh transporter (VAChT) knockdown homozygous (VAChT KD HOM ) mouse is a useful model for examining the cardiocirculatory sympathovagal balance. Therefore, we investigated whether cholinergic dysfunction caused by reduced VAChT expression could adversely impact hemodynamic parameter [arterial pressure (AP) and heart rate (HR)] daily oscillation, baroreflex sensi… Show more

“…The arterial baroreflex sensitivity was calculated using sequence method ( Bertinieri et al, 1988 ; Blaber et al, 1995 ) by using CardioSeries computer software V2.4 1 similar to other research in the literature ( Durand et al, 2015 ; Silva et al, 2015 ). Beat-to-beat RR intervals and SBP were input to the software, search for a sequence of at least three consecutive beats in which increase in SBP was followed by an increase in RR intervals (up slope) and decrease in SBP followed by a decrease in RR intervals (down slope) with a correlation greater than 0.8 was considered.…”

“…The slope of the linear regression lines between systolic SAP and PI was taken as a measure of spontaneous BRS. The baroreflex effectiveness index, which provides information on baroreflex function that is complementary to spontaneous BRS, was also calculated (31). It was defined as the ratio between the number of SAP ramps followed by respective reflex changes in PI and the total number of SAP ramps (independent of whether accompanied by the corresponding reflex PI ramps) observed over the time window studied.…”

“…The results from symbolic analysis demonstrated an increase in 2 LV pattern occurrence, with no changes in 0 V and 2 UV in ELA2KO mice. Notably, according to studies that used these nonlinear normalized indices, the 1 V pattern occurrence represents parasympathetic and sympathetic modulation, whereas the 2 UV pattern occurrence represents only parasympathetic modulation of the heart (Porta et al, 2007; Durand et al, 2015). Collectively, the data from spectral and symbolic analysis suggest that ELA2KO mice have a shift toward parasympathetic over sympathetic modulation of the heart, which likely contributed to the basal bradycardia exhibited by these mice.…”
