Automatic Measurement of Light Absorption and Fluorescence on Paper Chromatograms

Abstract: Previous automatic devices for scanning paper strip chromatograms have either been of the intermittent, nonrecording type or have required the use of an automatic recording spectrophotometer. The attachment described is designed for use with a commercially available monochromator employing a stabilized light source such as the Beckman Model DU spectrophotometer. An innovation in the assembly described is the use of interference filters to

“…In this case the lamp housing is replaced with one in which the lamp is removed and the solution may be inserted at the focal point of the mirror. Another modification of the Beckman Model DL* spectrophotometer places the fluorescent solution at the exit slit and a photomultiplier tube at the light entrance aperture (4%)• Brown and Marsh (14) use a modified Beckman Model DU to measure the fluorescence of paper chromatograms. A strip transporting mechanism and a chart recorder are among the attachments used; with a 1P28 tube a concentration of 2.6 y of riboflavin can be detected.…”

“…Data for the rutin standardization analyses are l)resented in figure 1 D 3 Agm. Ratios of the amount of rutin found to the amount actually applied varied from a low of 0.80 to a high of 1.10 (80 to 110 % recovery).…”

“…Many such methods have been described, all of which utilize a relation between amount of substance and some measure of zone dimension. Several devices have been used to measure the light absorption of substances on filter paper strips (3,4,14,15,16). In the present work modifications of two such techniques-those of Tennent, Whitla, and Florey (16) and of Paladini and Leloir (14)-have been applied to the estimation of rutin and four other buckwheat flavonoids.…”
