Automated multichannel fluorescent whole slide imaging and its application for cytometry

Abstract: Slide-based image cytometry (SBC) has several advantages over flow cytometry but it is not widely used because of its low throughput, complicated workflow, and high price. Fully automated microscopes became affordable with the advent of whole slide imaging (WSI) and they can be transformed into a cytometer. A MIRAX MIDI automated whole slide imager was used with metal-halide and light emitting diode (LED)-based fluorescent illumination, filter block changer, and a cooled monochrome charge coupled device camera… Show more

“…Cells in suspension are sucked or pressed into a cytometer apparatus by overpressure or the use of mechanical pumps. For analysis of solid tissues, slide-based cytometry is implemented by lamp illumination (Varga et al, 2009) and camera based detection, or laser excitation and fluorescence detection (Kamentsky et al, 1997). Solid tissue, however, cannot be analysed by flow cytometry microscopy (FCM) means without some form of initial tissue disintegration (Mittag and Tárnok, 2011).…”

“…Finally, advanced light emitting diode (LED) illumination has also been available in the recent past. Its remarkable advantages are the low working voltage and energy consumption, the long lifetime and the high speed which can significantly shorten the time of image capture (26,27).…”
