Automated Measurement of Pulmonary Emphysema and Small Airway Remodeling in Cigarette Smoke-exposed Mice

Abstract: COPD is projected to be the third most common cause of mortality world-wide by 2020 (1) . Animal models of COPD are used to identify molecules that contribute to the disease process and to test the efficacy of novel therapies for COPD. Researchers use a number of models of COPD employing different species including rodents, guinea-pigs, rabbits, and dogs (2) . However, the most widely-used model is that in which mice are exposed to cigarette smoke. Mice are an especially useful species in which to model COP… Show more

“…We excluded two mice in the Fam13a 1/1 air group and one mouse in Fam13a 2/2 CS group because of poor inflation for mean chord length analysis. Quality of lung inflation was judged by a senior investigator blinded to the experimental condition (26,27).…”

“…For each line of the grid projected upon the lung section image, the number of intercepts of the line with alveolar walls was counted. The average alveolar airspace size (represented by the mean linear intercept length or Lm, in μm) was then calculated by dividing the total grid length by the number of alveolar wall-grid line intersections as described previously (11,12).…”

“…For alveolar morphometric analysis, at least 15 images per mouse lung were randomly taken for analysis using methods described previously (22). Images were processed using Scion imaging software and analyzed for mean alveolar chord length (49).…”
