The kinetics and success of an industrial fermentation are dependent upon the health of the microorganism(s) responsible. Saccharomyces sp. are the most commonly used organisms in food and beverage production; consequently, many metrics of yeast health and stress have been previously correlated with morphological changes to fermentations kinetics. Many researchers and industries use machine vision to count yeast and assess health through dyes and image analysis. This study assessed known physical differences through automated image analysis taken throughout ongoing high stress fermentations at various temperatures (30 °C and 35 °C). Measured parameters included sugar consumption rate, number of yeast cells in suspension, yeast cross-sectional area, and vacuole cross-sectional area. The cell morphological properties were analyzed automatically using ImageJ software and validated using manual assessment. It was found that there were significant changes in cell area and ratio of vacuole to cell area over the fermentation. These changes were temperature dependent. The changes in morphology have implications for rates of cellular reactions and efficiency within industrial fermentation processes. The use of automated image analysis to quantify these parameters is possible using currently available systems and will provide additional tools to enhance our understanding of the fermentation process.

A Study of Nanosilver Colloid Prepared by Electrical Spark Discharge Method and Its Antifungal Control Benefits

This is a study of an antimicrobial test, including yeast, Aspergillus Niger, and Aspergillus Flavus, on a nanosilver colloid solution. The antibiosis is compared with a standard silver ion solution at the same concentration as in the experimental process. This study proved that the nanosilver colloid prepared by the electrical spark discharge method (ESDM) is free of any chemical additives, has a microbial control effect, and that the effect is much better than the Ag+ standard solution at the same concentration. 3M Count Plate (YM) is used to test and observe the colony counts. The microbial control test for yeast, Aspergillus Niger, and Aspergillus Flavus is implemented in the nanosilver colloid. In addition to Aspergillus flavus, an Ag+ concentration of 16 ppm is enough to inhibit the growth of the samples. At the same concentration, the nanosilver colloid has a much better microbial control effect than the Ag+ standard solution, which may be because the nanoparticle can release Ag+ continuously, so the solution using the ESDM has a more significant microbial control effect.

Staphylococcus aureus biofilms have been recognized as a leading cause of multiple infections, including implant-associated infections and chronic wounds. We evaluated the colonization capacity of two distinct textured prostheses by different bacterial strains. Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Proteus mirabilis and Enterococcus faecalis were evaluated. Initially, the hydrophobicity and biofilm formation capacity were determined. Subsequently, 20 fragments of vascular prosthesis and 20 silicone prostheses were embedded in suspensions with the microorganisms and incubated. The prostheses were then sown in culture medium and incubated for 48 hours. Petri dishes were photographed and analyzed by fractal dimension. The Kruskal-Wallis test and the Dunn test were applied for the analysis of biofilm formation. To compare the mean intensity for the type of bacteria and the type of prosthesis, a general linear model was applied. Staphylococcus aureus was the bacterium with the highest colonization density in both prostheses (p = 0.0001). E. coli showed strong adherence in the biofilm formation capacity test (p = 0.0001), however, it did not colonize either prosthesis. We demonstrated that Staphylococcus aureus has a greater affinity for vascular and silicone prostheses than other bacteria.
