Heat stress at reproductive stages of cotton crop is the main cause for instability in yield than other abiotic stresses of crops. An experiment was conducted to observe the impacts of heat stress at squaring and flowering stages of cotton and potential of exogenously applied ascorbic acid to alleviate this stress. The experiment was planted for two consecutive years, that is 2013 and 2014, in randomized complete block design with split plot arrangement having three replications. Treatments were comprised of heat stress in main plots, viz. H0 = no heat imposition; H1 = heat imposition at square initiation; H2 = heat imposition at flower initiation and foliar‐applied ascorbic acid in subplots, viz. A0 = control or water spray; A1 = 20 mg/L; A2 = 40 mg/L; and A3 = 60 mg/L. Results showed that heat stress badly affected plant water relations (relative water contents and osmotic potential), total chlorophyll contents and activities of antioxidant enzymes that led to a corresponding decrease in seed cotton yield. Foliar application of ascorbic acid (40–60 mg/L) ameliorated adverse effects of heat and thus helped the crop to improve its water relations, stay green and antioxidant enzymes resulting into better crop yield.

Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of bacterial blight (BB) of rice, uses transcription activator-like effectors (TALEs) to interact with the basal transcription factor gamma subunit OsTFIIAγ5 (Xa5) and activates the transcription of host genes. However, how OsTFIIAγ1, the other OsTFIIAγ protein, functions in the presence of TALEs remains unclear. In this study, we show that OsTFIIAγ1 plays a compensatory role in the absence of Xa5. The expression of OsTFIIAγ1, which is activated by TALE PthXo7, increases the expression of host genes targeted by avirulent and virulent TALEs. Defective OsTFIIAγ1 rice lines show reduced expression of the TALE-targeted susceptibility (S) genes, OsSWEET11 and OsSWEET14, which results in increased BB resistance. Selected TALEs (PthXo1, AvrXa7 and AvrXa27) were evaluated for interactions with OsTFIIAγ1, Xa5 and xa5 (naturally occurring mutant form of Xa5) using biomolecular fluorescence complementation (BiFC) and microscale thermophoresis (MST). BiFC and MST demonstrated that the three TALEs bind Xa5 and OsTFIIAγ1 with a stronger affinity than xa5. These results provide insights into the complex roles of OsTFIIAγ1 and OsTFIIAγ5 in TALE-mediated host gene transcription.

SBP-box (Squamosa-promoter binding protein) genes are a type of plant-specific transcription factor and play important roles in plant growth, signal transduction, and stress response. However, little is known about the role of pepper SBP-box transcription factor genes in response to abiotic stress. Here, one of the pepper SBP-box gene, CaSBP12, was selected and isolated from pepper genome database in our previous study. The CaSBP12 gene was induced under salt stress. Silencing the CaSBP12 gene enhanced pepper plant tolerance to salt stress. The accumulation of reactive oxygen species (ROS) of the detached leaves of CaSBP12-silenced plants was significantly lower than that of control plants. Besides, the Na+, malondialdehyde content, and conductivity were significantly increased in control plants than that in the CaSBP12-silenced plants. In addition, the CaSBP12 over-expressed Nicotiana benthamiana plants were more susceptible to salt stress with higher damage severity index percentage and accumulation of ROS as compared to the wild-type. These results indicated that CaSBP12 negatively regulates salt stress tolerance in pepper may relate to ROS signaling cascades.
