Autologous Serum For Expansion Of Human Bone Marrow Derivated Mesenchymal Stem Cell

MKH'ler %10 fetal bovin serumlu (FBS) besiyerinde üretilirler. Bu çal mada amaç FBS'un olas risklerinden uzaklalmas için otolog serum kullanarak klinik kullan m için uygun MKH üretimini sa lamakt r. Gereç ve Yöntemler:Ya lar 28-34 aras nda de i en 10 kemik ili i donoründen yaz l onamlar al narak 10 mL kemik ili i ve 500 mL periferik kan örne i al nd . Kemik ili inden fikol yo unluk derecelendirme yöntemiy-le mononükleer hücreler izole edildi. E it say da hücre (1x10 6 /mL) RPMI-1640 medium kullan larak %10 FBS, otolog serum (OS), %10, %5, %3 ve %1 konsantrasyonlar nda kültüre edildi. Her pasaj aras nda morfolojik de erlendirme, hücre say m ve ak m sitometrik yöntem-ler kullan larak immunfenotipik analizler yap ld . Üçüncü pasaja ula an gruplarda ya dokusuna farkl la t rma gerçekle tirildi. Bulgular:Hücre say m ve izolasyon aç s ndan en iyi grubun %10 OS grubu oldu u gösterildi. Buna ilave olarak %5 OS ve %10 FBS gruplar ndaki sonuçlar n benzer oldu u gözlemlendi. Ak m sitometrik sonuçlar de erlendirildi inde gruplar aras nda istatistiksel fark yoktu. Ya dokusuna farkl la t rma aç s ndan %3 OS ve %1 OS d nda di er gruplarda farkl la ma sa land .Sonuç: Sonuç olarak özellikle %10 OS ve %5 OS kullan larak MKH kültürleri yap labilece i saptand .Anahtar sözcükler: Mezenkimal kök hücre; otolog serum; kemik ili i; fetal buza serum.Objective: Mesenchymal stem cells (MSC), mostly derivated from bone marrow, are capable of differentiating into many tissues. In the culture medium consisting of 10% fetal bovine serum (FBS), MSCs can grown easily. To obtain insights into their ex-vivo expansion, the role of the autologous serum (OS) on growth and differentiation capacity of human bone marrow MSCs in comparison to cells grown in FBS was studied. Material and Methods:MSCs were cultured by using a RPMI-1640 medium consisting of 10% FBS, and OS 10%, 5%, 3% and 1%. Equally, 1x10 6 /mL cells were incubated into each flask. Cell count, viability, and flow cytometry studies were done at every passage after completing detachment in the cultures.Results: 10% OS group was found to be superior compared to the other study groups as regard to cell count and isolation. In addition, it has been observed that similar resuts were found in 5% OS and 10% FBS groups. Immunophenotyping analysis of the cells revealed no statistical difference between the groups. MSCs isolated from all study groups were successfully differentiated into adipogenic lineages except 3% and 1% OS groups. Conclusion:The expansion potency from bone marrowderived MSCs was maintained in culture media using 10% and 5% OS.
