Autofocusing MALDI mass spectrometry imaging of tissue sections and 3D chemical topography of nonflat surfaces

Abstract: We describe an atmospheric pressure matrix-assisted laser desorption-ionization mass spectrometry imaging system that uses long-distance laser triangulation on a micrometer scale to simultaneously obtain topographic and molecular information from 3D surfaces. We studied the topographic distribution of compounds on irregular 3D surfaces of plants and parasites, and we imaged nonplanar tissue sections with high lateral resolution, thereby eliminating height-related signal artifacts.

“…The fluorescent‐tagged antibody specific to proteins has been the major tool for localization studies . Recently, the integration of mass spectrometry with imaging has given rise to a new field of mass spectrometry imaging (MSI) . Utilizing MSI, various types of biomolecules, including proteins and lipids, can be mapped over the surface being visualized under the microscope.…”

“…Utilizing MSI, various types of biomolecules, including proteins and lipids, can be mapped over the surface being visualized under the microscope. Till now, major advancements have been in the profiling‐based approach with a combination of microscopy and matrix‐associated laser desorption and ionization (MALDI) or secondary ionization mass spectrometry (SIMS) . An integration of microscopy and MRM is a promising combination for the measurement of a specific set of proteins on the histopathological tissue of a diseased condition.…”

“…[10] Furthermore,a tmospheric near-field laser ablation coupled with MS has achieved al ateral resolution of 200 nm for easy-to-vaporize compounds. [12] Recently,a na utofocusing setup has been introduced for measuring the 3D chemical topography of non-flat sample surfaces, but only micrometerscale height variations can be resolved. This comprehensive information facilitates the visualization of 3D topographically reconstructed distributions of chemical species in subcellular regions.…”

[12] Recently,a na utofocusing setup has been introduced for measuring the 3D chemical topography of non-flat sample surfaces, but only micrometerscale height variations can be resolved. [12b] No single analytical technique is available that can provide the desired multimodal characterization abilities for topographically reconstructed single-cell imaging at submicrometer-scale lateral resolution.…”
