Autoacetylation of the Histone Acetyltransferase Rtt109

Abstract: Rtt109 is a yeast histone acetyltransferase (HAT) that associates with histone chaperones Asf1 and Vps75 to acetylate H3K56, H3K9, and H3K27 and is important in DNA replication and maintaining genomic integrity. Recently, mass spectrometry and structural studies of Rtt109 have shown that active site residue Lys-290 is acetylated. However, the functional role of this modification and how the acetyl group is added to Lys-290 was unclear. Here, we examined the mechanism of Lys-290 acetylation and found that Rtt10… Show more

“…The majority of HATs have been reported to have autoacetylation, such as p300 (25,26), Rtt109 (27), pCAF (28), human males absent on the first (hMOF) (29,30), yEsa1 (29), and TIP60. Autoacetylation is considered to be an important regulatory model for HATs.…”

“…In a small number of cases the precise contributions of specific, stoichiometric protein acetylation events have been characterized in detail with the authentic post-translational modification. Prior examples include cyclophilin (50), histone modifications (51-53), p53 (54), and acetyltransferase autoacetylation (55)(56)(57). By using expressed protein ligation on SAHH, we establish here, for the first time to our knowledge, the high resolution structural and functional consequences of acetylation at two independent sites in the same protein.…”

“…PCAF, the MYST family, p300, and Rtt109 are all known to be regulated by lysine autoacetylation, either through chemical or enzymatic means (12)(13)(14)(15)(16)(17)(18)(19). Interestingly, despite the commonality of this automodification among the acetyltransferases, the mechanism of regulation appears to vary between the proteins (20).…”
