AURKB, CHEK1 and NEK2 as the Potential Target Proteins of Scutellaria barbata on Hepatocellular Carcinoma: An Integrated Bioinformatics Analysis

Abstract: Objective: We aim to explore the potential anti-HCC mechanism of Scutellaria barbata through integrated bioinformatics analysis. Methods: We searched active ingredients and related targets of Scutellaria barbata via TCMSP database, PubChem and SwissTargetPrediction database. Then, we identified HCC disease targets from GEO dataset by WGCNA. Next, the intersected targets of disease targets and drug targets were input into STRING database to construct PPI networking in order to obtain potential therapeutic targe… Show more

“…The results based on hallmark gene sets revealed that the signaling pathways involved in NEK2 regulation of esophageal cancer growth include G2/M checkpoint, MYC, DNA repair, and MTORC1 signaling (Figure 3D). In summary, consistent with these reported [34][35][36], our results showed that NEK2 mainly acts on G2M checkpoint, RB, cell cycle, DNA replication, cell cycle G1/S phase transition, VGEF, MTOR, MYC, DNA repair, p53 signaling pathway, KRAS pathway, and MTORC1 (P < 0.05) (Figure 3E). More importantly, we found for the first time that NEK2 is associated with circadian pathways (Figure 3E).…”

“…Scutellaria barbata D.Don medicine trait is pungent, bitter, and cold. Its efficacy mainly acts on the lung, liver, kidney, and large intestine, and it can clear heat and detoxify, promote blood circulation and remove blood stasis, reduce swelling, and relieve pain [ 38 , 39 ]. Hedyotis diffusa Willd and Scutellaria barbata D.Don , when used in combination as a medicinal pair, exhibited anti-tumor effects.…”

“…In the current study, the results showed AURKB and FXOM1 gene are target genes for has-miR-204-5p with a downregulated expression in tumor samples. Aurora kinase B (AURKB) is a conserved family of serine/threonine kinase that has an essential function during cell cycle, such as chromosome segregation, spindle organization and cytokinesis [21,22]. AURKB is overexpressed in several cancers, such as triple-negative breast cancer.…”
