Aureonitol Analogues and Orsellinic Acid Esters Isolated from Chaetomium elatum and Their Antineuroinflammatory Activity

Abstract: Overexpression of various pro-inflammatory factors in microglial cells tends to induce neurodegenerative diseases, for which there is no effective therapy available. Aureonitol (1) and seven analogues, including six previously undescribed [elatumenol A−F (2−4, 6−8, respectively)], along with two new orsellinic acid esters [elatumone A and B (9 and 10)], were isolated from Chaetomium elatum. The structures of the compounds were established through comprehensive analysis of spectroscopic data, including high-res… Show more

“…RAW264.7 macrophages were plated into 6-well plates overnight, pretreated with 2 ml medium containing DMSO (0.1%), dexamethasone (30 μM) or 1 at concentrations of 7.5, 15, and 30 μM for 1 h and incubated with LPS (1 μg/ml). The details of protein sample extraction and experimental procedures are described in our previous work ( Ju et al, 2021 ).…”

“…Microglia are resident CNS cells that comprise 10% of the CNS population. In addition, microglia execute immune surveillance and perform macrophage-like activities, including the production of proinflammatory factors, such as interleukin (IL)-1β, interleukin-6 (IL-6) and tumor necrosis factor (TNF)-α, chemokines, small molecule messengers, such as nitric oxide, and reactive oxygen species (ROS) [ 1 , 3 , 4 ] . Uncontrolled and overactivated microglia can lead to pathological changes in the CNS and further lead to neurodegenerative diseases, such as Alzheimer’s disease and Parkinson’s disease [ 5 8 ] .…”
