Attenuated IL-2R signaling in CD4 memory T cells of T1D subjects is intrinsic and dependent on activation state

Abstract: The IL-2/IL-2R pathway is implicated in type 1 diabetes (T1D). While its role in regulatory T cell (Treg) biology is well characterized, mechanisms that influence IL-2 responses in effector T cells (Teff) are less well understood. We compared IL-2 responses in 95 healthy control and 98 T1D subjects. In T1D, low IL-2 responsiveness was most pronounced in memory Teff. Unlike Treg, CD25 expression did not influence the Teff responses. Reduced IL-2 responses in memory Teff were not rescued by resting, remained low… Show more

“…The sum of knowledge generated regarding T1D pathogenesis through particular HLA-DR alleles binding to specific autoantigenic epitopes could lead to the design of antigenspecific interventions (perhaps coupled with nonspecific ones) either in the pre-or in the new-onset T1D phase. The targets would probably have to include effector as well as regulatory CD4/CD8 T cells (48)(49)(50)(51)(52).…”

“…We and others have reported reduced IL-2 response in CD4 + CD45RO + memory T cells associated with T1D and T1Dassociated risk alleles (18,19,40,41). In this study, we assessed whether this phenotype precedes diagnosis in our risk-stratified cohort.…”

“…Tfh cells are also increased in autoantibody-positive (autoAb + ) children with impaired glucose tolerance, suggesting the evolution of a pathogenic Tfh population poised to promote B-cell responses during disease progression (15). Functional consequences likely underlie these phenotypes, as CD4 + Teffs are resistant to regulatory T-cell (Treg) suppression (16,17) and exhibit altered responses to cytokines, having a blunted response to interleukin (IL)-2 (18,19) and an enhanced response to IL-6 (20), in established T1D.…”
