Attempts to Mimic Docking Processes of the Immune System: Recognition-Induced Formation of Protein Multilayers

Abstract: The assemblage of protein multilayers induced by molecular recognition, as seen, for example, in the immune cascade, has been mimicked by using streptavidin as a docking matrix. For these experiments, this protein matrix was organized on liposomes, monolayers at the air-water interface, and self-assembled layers on gold, all three containing biotin lipids. The docking of streptavidin to biotin at liposomal surfaces was confirmed by circular dichroism. Mixed double and triple layers of streptavidin, concanavali… Show more

“…The strong interaction between biotin and streptavidin groups has been used for LbL multilayer assemblies in 1993. 141,142 Interactions between biotin and avidin, 143 sugar and lectin, 144,145 or antigen and antibody 146 have also successfully lead to preparation of LbL films. The wide range of noncovalent interactions useful for performing the LbL process has resulted in an incredibly wide variety of systems.…”

“…Some types of self-assembly and self-organization are achieved by covalent bonding of sulfur-bearing compounds on zero-valent metals such as gold, silver, platinum, and copper [1][2][3][4][5]. The controlled construction of ordered supermolecular organic structures on metal surfaces will help our understanding of twodimensional systems, such as biomembranes that play important roles in molecular recognition, signal transduction, and signal processing which are essential for cellular functions.…”

“…Highly stable molecular layers prepared by the step-bystep self-assembly method were used for developing hybrid devices [46,47] as illustrated schematically in figure 1(a). First, the commercially available microcantilever was cleaned using piranha solution (a mixture of 3:7 v/v of 30% H 2 O 2 and 98% H 2 SO 4 ) for 25 min, and rinsed with double distilled water three times for 5 min, followed by drying under clean air.…”
