Attachment of indium-111 labelled endothelial cells to pretreated polytetrafluoroethylene vascular grafts

Abstract: This study investigated the effect of different surface coatings on endothelial cell attachment to polytetrafluoroethylene (PTFE) vascular grafts. Small segments of PTFE vascular grafts were precoated with one of a number of substances: gelatin, poly-l-lysin, fibronectin, or collagen type I, III or IV. Indium-111 labelled endothelial cells were then seeded on to the grafts and left for either 10, 30, 60 or 120 min. The unattached cells were removed and the degree of cell attachment was calculated. All coatings… Show more

“…Collagen: Tissue engineering with collagen shows up to three times more cellular attachment than native non-coated grafts (BUDD et al, 1989;HAEGERSTRAND et al, 1993;THOMSON et al, 1989;VOHRA et al, 1991). This result was not confirmed by others (SANK et al, 1992;STANSBY et al, 1994).…”

“…It is known that an untreated ePTFE sur face has a low attachment of cells [7,8], Coat ing with laminin or fibronectin has altered this [7,8,11,12], but a preclotting process also enhances attachment [7,9] and is simple to accomplish. Fibronectin has the disadvan tage of being a favorite protein for staphylo coccus adherence [13,14], In spite of the pre clotting procedure, the attachment is much lower than that of preclotted Dacron.…”

“…The vein repeatedly was flushed with Minimal Essential Medium (MEM, GIBCO BRL, Izasa, Madrid) under sterile conditions to remove red blood cells. It then was filled to distention with a 0.1% CLS collagenase type I solution in 15 mmol/L CaCl 2 (Lorne Laboratories Ltd., Twyford, U.K.) following the technique described by Budd et al 14 The resulting cell suspension was centrifuged at 200 g for 7 min and the supernatant discarded. The cell pellet was resuspended in complete M-199 medium consisting of standard M-199 (GIBCO BRL, Izasa, Madrid), 20% fetal bovine serum (GIBCO BRL, Izasa, Madrid), 90 g/mL heparin-porcine intestinal mucosa (SIGMA, Madrid), 2 mmol L-glutamine (GIBCO BRL, Izasa, Madrid), 20 g/mL endothelial growth factor (SIGMA, Madrid, Spain), 20 nmol Hepes buffer (GIBCO BRL, Izasa, Madrid), and 10,000 IU/ mL penicillin plus 10,000 g/mL streptomycin (GIBCO BRL, Izasa, Madrid).…”
