Atrial Natriuretic Peptide Attenuates Colitis via Inhibition of the cGAS-STING Pathway in Colonic Epithelial Cells

Abstract: Atrial Natriuretic Peptide (ANP) has known anti-inflammatory effects. However, the role of ANP in Ulcerative colitis (UC) remains unclear. This study aimed to explore the expression and function of ANP in UC, and its potential regulatory role in the stimulator of interferon genes (STING) pathway. Human colon biopsy and serum samples were collected between September 2018 and December 2019 at Wuhan Union Hospital. Levels of ANP and its receptors and STING pathway components were detected in people with UC and mi… Show more

“…A very recent study has reported that decreased levels of ANP and NPR1 exacerbate ulcerative colitis in humans and the experimental mice. Mechanistically, this inflammatory response is mediated by ANP stimulator of interferon gene (STING) cascade in the process of colitis [ 15 ].…”

“…Natriuretic peptide receptor 1 (NPR1) (also known as NPRA or GC-A) is a membrane-bound receptor for atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP), which is expressed in many cell types and tissues including heart, vasculature, spleen, and gut [13][14][15]. Upon activation by ANP or BNP, NPR1 stimulates the production of cyclic guanosine monophosphate (cGMP) that promotes vascular tone and volume homeostasis by activation of protein kinase cGMP-dependent 1 (PKG), thereby controlling blood pressure and maintaining cardiovascular homeostasis [16,17].…”

“…Ten-week-old adult male C57BL/6 mice (25 ± 2 g) were obtained from the Animal Center of Lanzhou University (Lanzhou, China, SCXK (GAN) 2018-0002). All mice were housed in a specific pathogen-free separate room at 22 ± 2 °C with a 12 h light–dark cycle and 40–60% relative humidity, and adapted for 5 days before the experiments ( Nagata et al, 2021 ; Wu et al, 2021 ; Chen et al, 2022 ). Standard food and water were freely available in this period.…”

“…STING-deficient mice suffer less from severe colitis, while STING agonist exacerbates dextran sulfate sodium-induced colonic damage and inflammation [ 237 ]. In addition, atrial natriuretic peptide is reported to repair the gut barrier through the inhibition of STING pathway phosphorylation in epithelial cells [ 238 ]. However, the effects of STING remain controversial [ 239 ].…”
