Atrial fibrillation and microRNAs

Abstract: Atrial fibrillation (AF) is the most common sustained arrhythmia, especially in the elderly, and has a significant genetic component. Recently, several independent investigators have demonstrated a functional role for small non-coding RNAs (microRNAs) in the pathophysiology of this cardiac arrhythmia. This report represents a systematic and updated appraisal of the main studies that established a mechanistic association between specific microRNAs and AF, focusing both on the regulation of electrical and struct… Show more

“…Nonetheless, studies on the best normalization strategy in cardiac tissue remain sparse252627, and, to the best of our knowledge, no previous study has been performed to identify appropriate reference genes in the human atrial tissue for the study of AF. The lack of a general consensus on the best normalization strategy in this context has significant impact, considering the large increase in the number of studies analyzing the relationship between miRNA and AF in the last five years789. A variety of normalization strategies has been applied in different studies, which however were not validated in the specific experimental context of application2829.…”

“…AF is a multi-factorial disease, supported by altered electrophysiological and structural factors6. A growing body of works suggests miRNAs to act as potential mediators in the electrophysiological and structural remodeling of the atria maintaining AF789.…”

“…The prevalence of atrial fibrillation reaches values of 17.8% in people >85 years [1, 67, 68]. The development of a non-regular pattern of electrical activity might have detrimental consequences in hearts that are relatively stiff and relax slowly [1, 69, 70].…”

“…In addition, recent studies showed the microRNAs might play important roles in the adaptive process of the failing heart and affect the treatment response of cardiac arrhythmia via electrical membrane signaling and ion channel activation. [4244] A previous study also reported that metabolic syndrome was associated with poor outcome after PVC ablation. [45] This result emphasizes the role of metabolic, inflammatory, and redox processes, and ionic channel conduction properties on cardiac arrhythmia.…”
