ATR-FTIR spectroscopy and spectroscopic imaging for the analysis of biopharmaceuticals

Abstract: Attenuated Total Reflection Fourier Transform Infrared (ATR-FTIR) spectroscopy is a label-free, non-destructive technique that can be applied to a vast range of biological applications, from imaging cancer tissues and live cells, to determining protein content and protein secondary structure composition. This review summarises the recent advances in applications of ATR-FTIR spectroscopy to biopharmaceuticals, the application of this technique to biosimilars, and the current uses of FTIR spectroscopy in biophar… Show more

“…Among these three regions, MIR is the most widely applied for authentication analysis of fats and oils; therefore, in this review, we focused only on the MIR region. Recently, some reviews on the application of FTIR spectroscopy combined with chemometrics existed in different fields including biopharmaceuticals [ 35 ], halal food authentication analysis [ 36 ], authentication of meat and meat-based food [ 37 ], discrimination and authentication of herbal products [ 38 ], and authentication analysis of biomolecules in biomedical fluids [ 39 ]. FTIR spectra obtained during authentication analysis are complex and difficult to interpret; fortunately, with the development of chemometrics software and computer technology, the problems can be solved.…”

“…PLS-R using 2nd derivative spectra at optimized wavenumber regions of 1260–1080 cm −1 showed the best results for prediction of adulterant (FMO) levels providing the highest R 2 (0.999) with a residual predictive deviation (RPD) value of 31.91, low RMSEC of 0.53% v / v , and relative prediction error of 3.37%. The developed method could detect the adulteration level as low as 0.5% v / v [ 35 ].…”

“…In the case of thick or strongly absorbing SD samples, attenuated total reflectance FTIR (ATR-FTIR) spectroscopy, which has been utilized widely as a powerful technique in recent SD studies and the analysis of biopharmaceuticals, is recommended to overcome the intense peaks that are produced by these samples [ 67 , 68 , 69 , 70 ]. This system consists of an FTIR spectroscope and an ATR accessory to create an evanescent wave that is then directed into the sample [ 71 ].…”

Hannah Tiernan applied ATR-FTIR spectroscopy to determining protein content and protein secondary structure composition [15]. Li Zhihui employed ATR-FTIR spectra coupling with SVM classifier to identify graded asphalt [16].…”
