ATP serves an anti-inflammatory role by enhancing β-defensin-2 response in acute pneumonia of rat

Abstract: The aim of the current study was to evaluate the effect of ATP on the expression of rat β-defensin-2 (rBD-2) in a time-dependent manner, as well as its therapeutic value in an acute pneumonia rat model. A total of 30 rats as a treatment group and 30 as a control group were treated with the same dose of ATP and normal saline, respectively, lung tissues were isolated from rat and expression of rBD-2 mRNA was assessed with reverse transcription-quantitative polymerase chain reaction (RT-qPCR) at 12, 24 and 36 h f… Show more

“… 10 Recently, it was hypothesized that HBD-2 could suppress dendritic cell-mediated secretions of pro-inflammatory cytokines such as IL-1β, IL-12, and TNF-α in Inflammatory Bowel Disease (IBD) 28 as well as decrease IL-6 and TNF-α in lung tissues. 29 Also, HBD-3 reduces the secretion of IL-6 and IL-8, showing hopeful potential as adjuvant therapy for the treatment of inflammatory periodontitis. 9 …”

“…the mice showed ameliorated survival after skin flap infection by PA introduced through catheters (Casal et al ., 2019 ). Likewise, the overexpression of β‐defensin 2 in rats improved the outcome of PA lung infection, demonstrating the important protection exerted by these CAMPs in vivo (Shen et al ., 2017 ). As recently demonstrated for diverse HNPs (Zheng et al ., 2018 ), besides their obviously beneficial bactericidal activity (McHugh et al ., 2019 ), defensins show pro‐inflammatory properties which, depending on their levels and context of infection, can lead to excessive and harmful consequences for the host.…”

“…β-defensins are ubiquitously expressed by epithelial cells throughout the body, but β-defensin-1, -2, and -3 are the most studied respiratory tract β-defensins [ 116 , 176 ]. In addition to their antimicrobial role, β-defensins have been found to serve as both proinflammatory mediators [ 177 , 178 , 179 ] and attenuators [ 180 , 181 , 182 ] of the innate immune response, with the former better understood than the latter [ 183 ]. θ-defensins are not naturally expressed in humans and only exist in our genome as pseudogenes [ 116 ].…”
