A continuous process control apparatus for the determination of ATP in biomass was developed for monitoring activated sludge quality in biological wastewater treatment plants. The determination of ATP was based on the firefly luciferase reaction. The extraction efficiency of ATP removal from the biomass, influence of extractant on bioluminescence and potential for automation were tested during optimisation procedures. During the operation of a laboratory scale bioreactor, the automatic ATP analysis system provided a rapid and reproducible monitor of the biomass. It was a sensitive detector of inhibitory effects in the bioreactor and proved an excellent ‘toxicity monitor’.
