ATP-binding cassette proteins BCRP, MRP1 and P-gp expression and localization in the human umbilical cord

Abstract: The umbilical cord is a direct conduit to the fetus, hence transporters could have roles in partitioning substances between the maternal-placental-fetal units. Here we determined the expression and localization of the ATP-Binding Cassette (ABC) transporters BCRP (ABCG2), P-gp (ABCB1) and MRP1 (ABCC1) in human umbilical cords. The mRNA for BCRP and MRP1 was detected in 25/25 samples, but P-gp was detected in only 5/25. ABC transporter mRNA expression relative to 18S was 25.6±0.3, 26.5±0.6 and 22.2±0.2 cycles for… Show more

“…Meanwhile, 231/DDP cell line had a lower intracellular DDP concentration compared with 231/wt cell line, indicating 231/DDPcell line had a higher excretive ability of DDP, which might explain its strong drug resistance. Moreover, expressions of three drug-resistant membrane proteins (MRP1, P-gP and BCRP) [ 19 , 20 ] were remarkably increased in 231/DDP cell line. Collectively, these might be the physiochemical and molecular basis of DDP-drug resistance mechanisms.…”

“…The ABC transporter genes are classified into seven subfamilies (A to G). Among these transporters, ABCB1 (P-glycoprotein/P-gp/MDR1), ABCC1 (multidrug resistance protein 1/MDR1), and ABCG2 (breast cancer resistance protein/BCRP) have been extensively reported in many studies [ 160 ]. The most common indicator of multidrug resistance is P-glycoprotein (P-gp), which is a product of the multidrug resistance gene (MDR1) and a 170 kDa transmembrane phosphoglycoprotein that functions as an ATP-dependent efflux pump [ 161 ].…”

“…ABCC1 expression has been shown in many species, and in human, ABCC1 mRNA is expressed in various tissues including placenta, liver, and kidney [ 12 , 13 ]. ABCC9 is a subunit of ATP-sensitive K + channel and is expressed in many tissues including myometrium, cardiac and skeletal muscle [ 14 ].…”
