Atorvastatin and Aspirin as Adjuvant Therapy in Patients with SARS-CoV-2 Infection: A structured summary of a study protocol for a randomised controlled trial

Abstract: Objectives To assess the impact of adding statin (atorvastatin) and/or aspirin on clinical deterioration in patients infected with SARS-CoV-2 who require hospitalisation. The safety of these drugs in COVID-19 patients will also be evaluated. Trial design This is a single-centre, prospective, four-arm parallel design, open-label, randomized control trial. Participants The study will be conducted at National Cancer Institute (NCI), Jhajjar, Haryana, which is a part of All India Institute of Medical Sciences … Show more

“…Statins are inexpensive, easily available, and are already in widespread use for treatment of cardiovascular disorders, which has created considerable interest in further exploration of their repurposing as a new treatment for managing the severity of COVID-19 infections [16][17][18][19][20][21]. Statins are known to reduce the risk of major cardiovascular events and to counteract inflammation, the immune response, and oxidative stress damage [22][23][24][25], which may reduce the risk of a more severe disease course and improve outcomes in COVID-19 patients.…”

“…Interestingly, fondaparinux and atorvastatin exhibited very low RMSD (<3Å on average) among all 20 RBD-drug complexes. Both fondaparinux and atorvastatin have been proposed in the COVID-19 treatment [ [49] , [50] , [51] ] but their binding interactions with RBD has not been reported. Fondaparinux is a highly polar molecule and contains five monomeric sugar units.…”

“…Whether these medications are also protective among individuals without underlying indications for taking them such as CVD or hypertension is less clear and merits further study. Several randomized trials are currently underway evaluating the use of these medications for treatment of COVID-19 [29][30][31].…”
