Atomic Force Microscopy Imaging Demonstrates that P2X2 Receptors Are Trimers but That P2X6 Receptor Subunits Do Not Oligomerize

Abstract: P2X receptors are cation-selective channels activated by extracellular ATP. The architecture of these receptors is still not completely clear. Here we have addressed this issue by both chemical cross-linking and direct imaging of individual receptors by atomic force microscopy (AFM). Cross-linking of the P2X 2 receptor produced higher order adducts, consistent with the presence of trimers. The mean molecular volume of the receptor determined by AFM (409 nm 3 ) also points to a trimeric structure. P2X 2 recepto… Show more

“…This angle profile, with two peaks at around 90°and 180°, indicates that the Kv7 heteromer is indeed a tetramer. In contrast, a trimer would give a single angle peak at 120° (17), whereas a pentamer would give equally sized peaks at 72°and 144° (18).…”

“…We have developed a method, on the basis of AFM 4 imaging, for determining the arrangement of subunits within multimeric proteins (17)(18)(19). The method involves engineering specific epitope tags onto each subunit and expressing the proteins in a suitable cell line (e.g.…”

“…We have now used AFM imaging of Ab-decorated receptors to provide information about the architecture of three types of ligand-gated ion channel, the GABA A (24), the P2X (18), and the 5-HT 3 receptors. We suggest that the method that we describe here can be applied not only to other members of the ligand-gated ion channel superfamilies (1-3, 39) but also more widely to other types of multisubunit protein.…”

“…The solubilization͞purification procedure was done as described in ref. 18 for P2X receptors. Briefly, a crude membrane fraction prepared from transfected tsA 201 cells was solubilized in 1% (wt͞vol) 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate, and the solubilized material was incubated with Ni 2ϩ -agarose beads (Probond, Invitrogen).…”
