AtHMA1 Is a Thapsigargin-sensitive Ca2+/Heavy Metal Pump

Abstract: The Arabidopsis thaliana AtHMA1 protein is a member of the P IB -ATPase family, which is implicated in heavy metal transport. However, sequence analysis reveals that AtHMA1 possesses a predicted stalk segment present in SERCA (sarcoplas- Plant Ca 2ϩ and heavy metal-ATPases belong to the superfamily of P-ATPases (1, 2). Their common characteristic is the presence of a phosphorylated intermediary in the catalytic cycle. In Arabidopsis, the heavy metal-ATPases belong to the P IB subfamily and normally have eight … Show more

“…It is interesting to note that thapsigargin inhibited the ER-calcium ATPase in U. compressa as it has been shown mammalian cells. 17 These results contrast with data obtained in terrestrial plants where the ER-calciumATPase is not inhibited by thapsigargin, with the exception of a heavy metal/ calcium ATPase detected in the Golgi apparatus of pea seedlings 15 and a similar enzyme recently cloned in Arabidopsis thaliana. 16 In addition, time required to reach the maximal level of calcium depletion induced by thapsigargin i.e., 45 min is similar to that observed in mammalian cells, i.e., 30 min.…”

“…Members of the P 1B4 subgroup include broad-specificity transporters of metals. A. thaliana HMA1 was shown to be a Ca 2ϩ /heavy metal pump localized in the chloroplast envelope, which delivers copper from the cytosol into the chloroplast stroma and detoxifies chloroplasts from Zn 2ϩ under zinc excess (8,9). A similar protein from barley (Hordeum vulgaris), HvHMA1, was shown to be involved in mobilization of zinc and copper from plastids under zinc or copper deficiency or from the intracellular compartments of the mineral storage site of grains (aleurone cells) during grain filling and germination (10).…”

“…At HMA1, a heavy metal transporter involved in detoxification processes for heavy metals, is a P I -ATPase that localizes to the chloroplast envelope. At HMA1 transports Ca 2+ and heavy metals, such as copper, with high affinity and, similar to Ca 2+ pumps from animals, is specifically inhibited by thapsigargin (Seigneurin-Berny et al, 2006;Moreno et al, 2008).…”
