Cannabis sativa L. is an annual herbaceous crop grown for the production of long extraxylary fibers, the bast fibers, rich in cellulose and used both in the textile and biocomposite sectors. Despite being herbaceous, hemp undergoes secondary growth and this is well exemplified by the hypocotyl. The hypocotyl was already shown to be a suitable model to study secondary growth in other herbaceous species, namely Arabidopsis thaliana and it shows an important practical advantage, i.e., elongation and radial thickening are temporally separated. This study focuses on the mechanisms marking the transition from primary to secondary growth in the hemp hypocotyl by analysing the suite of events accompanying vascular tissue and bast fiber development. Transcriptomics, imaging and quantification of phytohormones were carried out on four representative developmental stages (i.e., 6–9–15–20 days after sowing) to provide a comprehensive overview of the events associated with primary and secondary growth in hemp. This multidisciplinary approach provides cell wall-related snapshots of the growing hemp hypocotyl and identifies marker genes associated with the young (expansins, β-galactosidases, and transcription factors involved in light-related processes) and the older hypocotyl (secondary cell wall biosynthetic genes and transcription factors).

Although there are no known sources of genetic resistance, some Citrus spp. are reportedly tolerant to huanglongbing (HLB), presumably caused by 'Candidatus Liberibacter asiaticus'. Time-course transcriptional analysis of tolerant rough lemon (Citrus jambhiri) and susceptible sweet orange (C. sinensis) in response to 'Ca. L. asiaticus' infection showed more genes differentially expressed in HLB-affected rough lemon than sweet orange at early stages but substantially fewer at late time points, possibly a critical factor underlying differences in sensitivity to 'Ca. L. asiaticus'. Pathway analysis revealed that stress responses were distinctively modulated in rough lemon and sweet orange. Although microscopic changes (e.g., callose deposition in sieve elements and phloem cell collapse) were found in both infected species, remarkably, phloem transport activity in midribs of source leaves in rough lemon was much less affected by HLB than in sweet orange. The difference in phloem cell transport activities is also implicated in the differential sensitivity to HLB between the two species. The results potentially lead to identification of key genes and the genetic mechanism in rough lemon to restrain disease development and maintain (or recover) phloem transport activity. These potential candidate genes may be used for improving citrus tolerance (or even resistance) to HLB by genetic engineering.

Arabinogalactan-proteins (AGPs) are present on the surface of all plant cells. AtAGP17, 18 and 19 comprise the lysine-rich AGP subfamily in Arabidopsis and consist of an N-terminal signal peptide, a classical AGP domain interrupted by a small Lys-rich region and a C-terminal glycosylphosphatidylinositol (GPI) anchor addition sequence. Organ- and tissue-specific expression patterns and subcellular localization of AtAGP18 were studied and compared to other Lys-rich AGPs. AtAGP18 was highly expressed in roots, flowers and stems and weakly expressed in seedlings and rosettes. High AtAGP18 promoter activity was closely associated with vascular tissues and high in young organs as well as styles. Microarray and massively parallel signature sequencing (MPSS) data were also examined and showed largely consistent transcription profiles of AtAGP18. On the protein level, AtAGP18 was most abundant in roots and flowers, moderate in stems, seedlings and siliques and low in rosette leaves. Furthermore, AtAGP18 was localized to the plasma membrane and to Hechtian strands, following plasmolysis of tobacco cultured cells expressing a green fluorescence protein (GFP)-AtAGP18 fusion protein. Localization of AtAGP18 on the plasma membrane was further confirmed by biochemical two-phase fractionation and Western blotting. These expression and localization data further our understanding of AtAGP18 and provide a molecular basis to approach and decipher its function.
