Asymmetric synthesis of ß-amino acids and α-substituted β-amino acids

Abstract: Giuliana Cardillo was born in Anconu, Italy. She received her degree in Chemistrvfrom the University of Rome. Then she moved to the Politecnico of Milan working under Professor A. Quilico on naturally occurring chromenes. After two year5 at the University of Bari she moved to the Department of Chemistry 'G. Ciamician' of the Universiq of Bologna working with Profesmr G. Cainelli on the syntheJi5 of terpenoids. She became ProfeJJor of Organic Chemistry in 1980. Her major re5earch interest5 lie in the area of as… Show more

“…Haemoglobin is Fe(II) containing complex acts as a oxygen carrier. Ca(II) complexes is basic constituents of bone, Zn 2+ ions found in three-dimensional structural framework of proteins are good examples of metal complexes in biological systems 13 . Metal ions act as redox transfer and hence metal complexes showed medicinal applications like organic compounds which could be used in drug discovery efforts 14 .…”

“…These compounds have a great relevance since they are present in biologically active peptides, 7 and are also valuable for the preparation of peptidomimetics, 8 substituted β-lactams 9 and β-amino 45 acids. 10 The selective synthesis of these unprotected synthons is challenging, 11 and no general strategy comprising the direct asymmetric reductive amination of the α-substituted β-keto ester precursors has been described until now. 12 50 Scheme 2 Synthesis of racemic aliphatic α-substituted β-amino ester derivatives through enamine formation-reduction protocol.…”

“…To perform these biocatalyzed transformations, a series of 24 commercially available transaminases, Codex ® Transaminase 10 Screening Kit, was tried as most of them are able to work under the very convenient conditions using isopropylamine in molar excess as amino donor. 15 In a first set of experiments, unsubstituted ethyl acetoacetate 1a was studied as substrate for these TAs (Tables S2-S3 in ESI) using alanine or isopropylamine as amino 15 donors.…”

“…[1][2][3] Among these, approaches involving an aza-Michael reaction have often been used. In this process, the asymmetric induction may come either from a chiral nitrogen nucleophile, [4][5][6] or from a chiral conjugate electrophile, [7][8][9][10][11] or from a chiral ligand, employed either stoichiometrically 12,13 or catalytically.…”

“…In the absence of a suitable anomalous scatterer, the Friedel pairs have been merged, the absolute configuration of the unknown chiral centre being deduced from that of the known one. Crystal data for 3h (major isomer): C 21 H 21 NO 6 , M = 383.39, orthorhombic, space group P2 1 . CCDC-990814 contains the supplementary crystallographic data for this paper.…”
